Ethanamizuril,a novel anti-coccidiosis triazine compound,was synthwsized by the shanghai Veterinary Research Institude of the Chinese Academy of Agricultural Sciences,to study and compare its pharmacokinetics in healthy and infected coccidia chickens.This article established two detection methods for the determination of Ethanamizuril in chicken plasma and for the determination of Ethanamizuril in chicken cecum by HPLC,in addition,the blood pharmacokinetics parameters of Ethanamizuril in healthy chickens and infected chickens were compared using chicken plasma assays;Comparison of cecal pharmacokinetics parameters of Ethanamizuril in healthy chickens and chickens infected with coccidiosis using chicken cecal test,provides reference for the clinical administration of Ethanamizuril.Determination of plasma anticoccidial drug Ethanamizuril in HPLC:plasma samples wereprecipitated by methanol,C 18 columnseparation,detection at251 nm wavelength,internal standard method for quantification.Samaplatril has a good linear relationship in the range of 0.05-20?g/mL?R2=0.999?.The HPLC method for the determination of Ethanamizuril content in chicken cecum was performed.After the chicken cecal sample was homogenized,it was extracted with acetonitrile and hexane was degreasing.The results showed that the method has a good linear relationship in the detection of Ethanamizuril in the cecum of chicken 0.20-0.4mg/kg?R2=0.994?.Pharmacokinetic experiment results of Ethanamizuril in healthy and infected cecum of chickens:In the healthy group,the Tmax was 5.125 h,the Cmax was 9.964?g/mL,the t1/2was 15.558 h,the AUC0-t-t was 199.824 mg/L*h,and the MRT0-t-t was 17.612 h;In the infective coccidia group,the Tmax was 5.25 h,Cmax was 8.733?g/mL,t1/2/2 was 15.971 h,AUC0-t-t was169.587 mg/L*h,MRT0-t-t was 17.745 h.Comparing the curve and pharmacokinetic parameters of Ethanamizuril in health and infection with coccidiosis,it was found that there was no significant difference in blood pharmacokinetics,this proves that the blood pharmacokinetics of Ethanamizuril in chickens will not be affected by whether chickens are infected with Eimeria tenella.Pharmacokinetic experiment results of Ethanamizuril in healthy and infected cecum of chickens:In the healthy group,the Tmax was 6.125 h,Cmax was 0.696 mg/mL,t1/2/2 was128.744 h,AUC0-t-t was 18.421 mg/L*h,MRT0-t-t was 42.238 h;In the infective coccidia group,the Tmax was 7.5 h,the Cmax was 1.31 mg/mL,the t1/2/2 was 42.783 h,the AUC0-t-t was30.946 mg/L*h,and the MRT0-t-t was 32.491 h.Comparing the curve and pharmacokinetic parameters of Ethanamizuril in health and infection with coccidiosis cirrhosis,differences were found in the healthy and infective coccidia groups,the Tmax,Cmax,and AUC0-t-t of chickens infected with coccidia are larger than that of healthy chickens,but t1/2/2 is decreased,prove that the infection of coccidiosis can affect the distribution and elimination of Ethanamizuril in chicken cecum.In summary,the blood pharmacokinetics of Ethanamizuril in chickens was not affected by chicken-infected coccidia,but the pharmacokinetics of Ethanamizuril in chicken cecum was affected by chicken-infected coccidia,this provides experimental evidence for further studies of Ethanamizuril.
