| Dendrobium officinale is a perennial herb of Dendrobium,which has extremely high medicinal value and is reputed as "gold in medicine".Due to the small size of dendrobium officinale seeds and immature embryo development,the natural reproduction rate is low.Under natural conditions the growth and development can be completed with bacteria.Therefore,it is difficult to breed in the field or seedbed,so it is difficult to produce enough seedlings for cultivation.At present,the rapid development of dendrobium in the wild has been preliminarily completed,but there are still many problems to be solved.To accelerate the tin caulis dendrobii tissue rapid propagation technology more efficient rationalization,this experiment progress is to tin caulis dendrobii somaclone as research materials,research the combination of 6-different concentrations of BA and NAA tin caulis dendrobii original culture influence,growth regulator combination type and proportion of tin caulis dendrobii successive transfer culture influence,and different concentration of IBA on tin caulis dendrobii begins to take root,the influence of the rapid reproduction technology of tin caulis dendrobii has carried on the improvement of optimization.The results were as follows:1、Primary culture:The optimum medium for the primary culture of Dendrobium officinale was MS+ 6-BA1.0 mg/1-1 +NAA0.01 mg/1-1,the differentiation rate was 100%,and the proliferation multiple was 2.95.2、Subculture:The most suitable type of cytokinin in subculture of Dendrobium officinale was 6-BA,and the growth element was IBA.The optimum medium for the secondary culture of Dendrobium officinale was MS+ 6-BA0.5mg/1-1 + naa0.5mg/1-1,and the differentiation rate was 100%,and the value-added rate was 6.05.3、Rooting culture:It is most suitable for the rooting culture of Dendrobium officinale was 1/2MS+IBA1.0 mg/1-1,the rooting rate was 100%,the average rooting number was 3.6,the average of root length was 0.36cm,and the average of root was 0.08 cm. |
