‘Yuluxiangli' pear has good inner quality but its low rate of calyx-abscission,irregular irregularities and fruit surface protrusions affect its appearance quality.For this reason,promoting the shedding of fruit sepals and improving their internal and external quality is an urgent problem to be solved in production.This study investigated the effects of pollinated varieties,tree vigor,moisture treatment,fertilization scheme and spraying different concentrations of PBO on the fruit shape and quality of 'Yuluxiangli' Pear.RNA-Sep expression profiling techniques based on high-throughput sequencing were used to analyze genes with significant differences in expression of pear fruits under smooth and matte surfaces.The main conclusions are as follows:1.‘Yuluxiangli' had the best fruit shape with 'Huangguan' as the pollination variety.The roundness value was 1.05 and the declination rate was 15.06%.Both of them were significantly lower than other treatments.There was no significant difference among other pollination varieties.There was no significant difference in the overall quality and taste after treatment with different pollinated varieties.The pollinated variety with the lowest crispy pulp was 'Huangguan' and followed by 'Xuehuali'.2.The calyx abscission rate of fruit with strong vigor was only 5.82%.With the decrease of tree vigor,the delocation rate increased significantly.The rate of calyx abscission from the moderate tree vigor was 3-4 times of the strong tree.The weak or moderate fruit surface raised level was significantly lower than that of strong and stronger tree vigor and fruit shape was better.In addition,poor or too strong tree vigor will cause serious fruit skewness and reduce the content of soluble solids in the fruit.3.Compared with single fertilizer,the application of different fann fertilizers can increase the fruit calyx abscission rate and reduce the fruit shape index and the degree of fruit surface bulge.All of them reached a significant level and calyx abscission rate with cow was 5.8 times as high as that of the control,which was significantly higher than that of chicken manure and sheep dung treatment.At the same time,the content of organic matter in soil could improve effectively compared with the application of different farmyard manure and fertilizer.The content of soil available phosphorus was significantly increase by 79.49%with the application of chicken manure and both chicken manure and sheep manure can be significantly improved the content of alkali hydrolysable nitrogen and available potassium.4.PBO had great influence on the 'Yuluxiangli' and the effect of improvement was significant.The best combination of internal and external quality was 1500 times PBO.The calyx abscission rate increased gradually with the increase of PBO concentration and all of them reached over 74%,which was significantly higher than that of the control group(22.03%).The single fruit weight,vertical and horizontal diameter,fruit shape index and the degree of bulging on the fruit surface showed a gradual decrease trend with the increase of PBO concentration.The fruit longitudinal diameter of 300 and 500 times PBO treatments was significantly lower than that of other treatments.The fruit shape index of 300 times PBO treatment was significantly lower than that of other treatments and it was only 0.94.The deviation rate and roundness of the treatments were significantly lower than that of the control except 2000 times PBO concentration.There were no significant changes in fruit pulp texture after PBO treatment.5.The calyx abscission rate with controlled irrigation was 2 times that of the irrigation treatment after flowering and fruiting and the difference was significant.The controlled irrigation was significantly reduced the deviation rate and the surface protruding degree compared with the control and improved the appearance quality of 'Yuluxiangli,.In addition,controlled treatment and spraying PBO could better maintain the quality and taste of4 Yuluxianglf but PBO treatment has a higher calyx abscission rate,which is twice that of water treatment.6.Transcriptome sequence analysis found that 40 differentially expressed genes were screened out,of which the expression of PBO treatment was up to 17,down 23.The gene DELLA related to gibberellin signal transduction and MYC2 gene involved in methyl jasmonate signal transduction respond to PBO processing.
