[Objective] A large number of references have shown that fluoride can cause liver damage.The rats were added different calcium that study expression of apoptosis-related proteins and genes.The calcium can alleviate fluorosis to determine the optimal calcium dose and provide theoretical basis.[Methods ] There are 40 male SD rats with an average weight of 120 g that were divided into five groups,control group(standard diet+ distilled water),fluoride group(standard diet + 150mg/l Na F water),fluoride + low calcium group(standard diet + 150mg/l Na F+o.5%Ca water),fluoride+ medium calcium group(standard diet + 150mg/l Na F + 1%Ca water),and fluoride + high calcium group(standard diet + 150mg/l Na F + 1.5%Ca water).During feeding,environment is suitable and feeding for 120 days freely.We observed the mental state and to record the weight and intake of rats.After 120 days,the blood was collected by eyeball from rats,and serum and liver were collected.Weighing liver quality and analyzing liver organ index.Serum for AST and ALT activity were determined with kits;HE was used to observe the pathological and structural changes of liver tissue;Immunohistochemistry was used to detect the expression of apoptosis protein Bax,Bcl-2,ROCK I,Cyto-C and BMP-2 in liver tissue.The effect of calcium on the liver chronic fluorosis was discussed in rats,and the apoptosis of rats' hepatocytes was analyzed.For prevention and treatment of chronic fluorosis,the calcium was determined to provides the optimal dose and a theoretical basis.[Results](1)The rats' mental state was normal,teeth were intact,and rats were sheen in the control group.Compared with the control group,the growth and development of the rats were worse in the fluoride group,the teeth were slightly yellowed and the hair was dimmed.The rats had better remission,development,tooth appearance and glossy coat in the fluoride + high calcium group.(2)There was no significant change in the liver organ index in each group,compared with the control group.(3)The fluoride group of ALT activity was significantly lower compared with the control group(p <0.01).Compared with the fluoride group,the activity of ALT in the fluoride + medium calcium group and fluoride + high calcium group was significantly lower(p < 0 05).The activity of AST in fluoride group was significantly higher than that in the control group(p < 0 01).However,the activity of AST in the fluoride + medium calcium group was significantly lower than that in the fluoride group(p < 0 05).(4)Compared with the control group,the activities of T-AOC,GSH-PX and SOD in the fluoride group decreased significantly(p < 0.05).The content of MDA in the fluoride group was significantly higher than that in the control group(p < 0.05).Compared with the fluoride group,the activities of T-AOC,MDA and SOD in the fluoride + medium calcium group and fluoride + high calcium group were significantly higher(p < 0 05);The expression levels of GSH-Px in the fluoride + medium calcium group was significantly higher(p < 0 05).(5)The results of HE staining showed that: Fluoride can cause damage to the morphology and structure of liver tissue,and the cells become granular degeneration,vacuole and necrosis.Calcium can relieve the bad phenomenon of tissue caused by sodium fluoride.(6)The results of immunohistochemistry showed that:The expression levels of Bax,Cyto-C,ROCK I and BMP-2 increased significantly(p <0.001),and the expression levels of Bcl-2 decreased significantly in compared to the control group(p <0.001).Compared with fluoride group,the expression of Bax and Cyto-C in fluoride + medium calcium group and fluoride + high calcium group decreased significantly(p < 0 05),but the expression level of Bcl-2 in calcium group increased significantly(p < 0 05),and the expression of ROCK I in fluoride+high calcium group decreased significantly(p < 0 01),and the expression level of BMP-2 in fluoride + high calcium group was significantly lower(p < 0 05).(7)The results of q RT-PCR showed that:Compared with the control group,the expression of Bax,ROCK I and Cyto-C in the fluoride group increased significantly(p <0.01),the Bcl-2 gene decreased significantly(p <0.01),the BMP-2 gene was significantly higher(p <0.05).The expression of Bax,ROCK I and Cyto-C in the fluoride + low calcium group and the fluoride + medium group decreased significantly(p <0.05),the Bcl-2 gene was significantly higher in the fluoride + medium calcium group and fluoride + high calcium group(p <0.01),but BMP-2 gene in the fluoride + medium calcium group was significantly decreased(p <0.05).[Conclusion] The result showed that: Fluoride can affect the growth performance and expression of liver-related apoptotic proteins and genes in rats,but calcium can relieve the symptoms.1% calcium is the best concentration to improve liver apoptosis in fluorosis rats.However,low calcium can aggravate hepatocyte apoptosis induced by fluorosis.
