The study explored the effects of microbe organic fertilizer on soil bacterial community structure,soil fertility and asparagus quality,which provided a theoretical basis for improving domestic farming patterns and improving the quality of asparagus planting.In this study,Bacillus megaterium,Bacillus subtilis,Bacillus coagulans and Pseudomonas fluorescens were mixed together by equal proportion in corn dry powder medium and fermented for 3 days after they respectively cultured in seed medium for 24 h.And the fermentation broth was mixed with wormcast in a ratio of 0.3:1(volume ratio)to manufacture a microbe organic fertilizer.The experiment set a compound fertilizer treatment and four dosages of microbe organic fertilizer were designed as base fertilizer and contained CK(no fertilizer).The experiment,in the experimental greenhouse,was completely randomized and sampled and tested at 30 days,60 days and 90 days,respectively.The analysis results are as follows:1.A total of 46,214 operational taxonomic units(OTUs)were obtained from the second-generation high-throughput sequencing of bacteria in soils at different times.The bacterial classification level chart showed that the proteobacteria had the highest abundance which accounted for 25.18%?60.00%in each treatment,followed by the Bacteroides and the thick-walled bacteria.At the genus classification level,the proportion of Pseudomonas was the highest in each treatment which accounted for 0.07%?9.81%,followed by Bacillus.In the microbe organic fertilizer treatment contained 400,600 and 800 g/m2 at 90 days the abundance of nitrogen-fixing bacteria was 1.29%,1.53%,and 1.68%,respectively.PCoA analysis showed that the soil bacterial community structure of the 600 g/m2 and 800 g/m2 microbe organic fertilizer treatment was more similar to the treatment of fertilization at 60 and 90 days,while the compound fertilizer treatment was quite different from remaining treatment.In the soil used the microbe organic fertilizers,Bacillus,Pseudomonas,etc.,as well as soil bacterial abundance and diversity had increased to varying degrees.The PCR-DGGE results similar to those of high-throughput sequencing,which found that it had higher bacterial diversity at each stage in microbe organic fertilizer treatment(800g/m2),and it was found that the content of Bacillus megaterium and Bacillus subtilis increased in the soil after used the microbe organic fertilizer2.The results of soil fertility factors showed that treatment with microbe organic fertilizers(600g/m2 treatment)had the best performance in the soil fertility factors except for the total nitrogen content at 30d,60d and 90d.And the effects of microbe organic fertilizer(600g/m2 treatment)on soil total nitrogen,available phosphorus and available potassium were the most obvious at 90d,which were 6.34%,37.31%and 70.49%respectively higher than the compound fertilizer treatment,and microbe organic fertilizer(800g/m2 treatment),affecting soil total phosphorus,total potassium,organic matter was the most obvious,compared with compound fertilizer respectively increased by 1.43%,5.82%,1 8.82%.Soil enzyme activity was the best in soil urease and cellulase in the fertilization treatment for 90d microbe organic fertilizer(800g/m2 treatment),which respectively increased by 347.78%and 106.58%compared with CK.It had the best effect on the activities of sucrase,catalase and phosphatase in microbe organic fertilizer(600g/m2 treatment),which were respectively 90.77%and 134.39%higher than CK.Compared with CK,the acidic,neutral and alkaline phosphatase respectively increased by 21.35%,40.83%and 35.97%.Soil quality was improved after used microbe organic fertilizer3.The results of asparagus quality index showed that the rutin content of the asparagus was the best in the microbe organic fertilizer(800g/m2 treatment),which respectively increased by 5.97%,18.38%and 12.07%compared with the compound fertilizer treatment.The Vc content respectively increased by 3.75%,11.43%,and 9.44%compared with the compound fertilizer treatment,but there was no significant difference in glutamic acid and aspartic acid of asparagus of each treatment.In summary,the study found that the application of the microbe organic fertilizer and compound fertilizer had significant activation of soil bacteria,which increased soil bacterial diversity and abundance,and significantly improved soil enzyme activity and soil fertility.In this experiment,the content of rutin and the content of Vc which was the main secondary metabolites of asparagus were significantly increased by changing the soil environment of asparagus,but its effect on glutamic acid and aspartic acid was small.The microbe organic fertilizer constructed in this study significantly improved the soil environment and significantly promoted the quality of asparagus.
