| Dictamnus dasycarpus Turcz.belongs to the perennianl herbaceous plants of Rutaceae Dictamnus,and its dried root bark is called Dictamni Cortex which is a traditional Chinese herbal medicine.Paeonia lactiflora Pall.is a perennial herbaceous plant of the Paeoniaceae Paeonia,and its dried root is the raw material of Paeoniae Radix Rubra.In this study,the seeds of Dictamnus dasycarpus Turcz.and Paeonia lactiflora Pall.were used as experimental materials,the seeds of Dictamnus dasycarpus Turcz.with water content of 4.2813.22%and the seeds of Paeonia lactiflora Pall.with water content of4.1010.07%were stored indoors,outdoors,in germplasm bank(short﹣term storage,medium﹣term storage,long﹣term storage),and changes of seed vigor were measured periodically,and selected suitable storage conditions for seeds.The relationship between seed vigor and physiological and biochemical indexes was explored through artificial seed aging test.Seed initiation and repair techniques were studied by using the artificially aged Dictamnus dasycarpus Turcz.seeds and Paeonia lactiflora Pall.seeds.It provided technical support for the conventional storage,germplasm bank preservation and the initiation and repair of the aged seeds,and it was of great significance to the production and germplasm conservation of Dictamnus dasycarpus Turcz.and Paeonia lactiflora Pall.The main research results are as follows:1.The viability of the seeds of Dictamnus dasycarpus Turcz.showed decreasing trends during the storage,after 12 months of storage,the viability of the seeds with a water content of 8.09%in the long-term storage decreased from 93.33%to 71.33%,the viability of the seeds under other storage conditions decreased to less than 60%.It was explained that the seeds of Dictamnus dasycarpus Turcz.were not resistant to store,long-term storage and 8.09%water content of seeds could prolong the life of the seeds.The vigor of the seeds of Paeonia lactiflora Pall.under different water content and storage conditions showed slow decreasing trends during the storage,after 12 months of storage,the viability and germination rate of the seeds were above 90.00%.It was explained that the seeds of Paeonia lactiflora Pall.were resistant to store.Preliminary results showed that the suitable water content of Paeonia lactiflora Pall.seeds in indoor,outdoor and germplasm bank was 4.1010.07%2.During the 12 months storage of Dictamnus dasycarpus Turcz.seeds under different storage conditions,the content of soluble sugar and protein displayed the tendency rising up at the beginning and declining up in the late,the content of MDA increased,and the activity of CAT,SOD and POD decreased.CAT and POD activity of Dictamnus dasycarpus Turcz.seeds with a water content of 8.09%decreased slightly during long-term storage,while MDA content increased slightly and the seed viability decreased slightly.During the 12 months storage of Paeonia lactiflora Pall.seeds under different storage conditions,the content of soluble sugar,and the activity of CAT,SOD and POD decreased slightly,the content of soluble protein displayed the tendency rising up at the beginning and declining up in the late and there was no significant change before and after storage,the content of MDA increased slightly,and the seeds remained high in vigor3.In the process of artificial seed aging of Dictamnus dasycarpus Turcz.seeds and Paeonia lactiflora Pall.seeds,the seed vigor,soluble sugar content,soluble protein content,SOD,POD and CAT activity of Dictamnus dasycarpus Turcz.and Paeonia lactiflora Pall.decreased,MDA content rised.The results of correlation analysis showed that the seed vigor of Dictamnus dasycarpus Turcz.and Paeonia lactiflora Pall.was positively correlated with soluble sugar content,soluble protein content,SOD,POD and CAT activity,but negatively correlated with MDA content.4.Four initiators were used to repair the artificial aging seeds of Dictamnus dasycarpus Turcz.and Paeonia lactiflora Pall.,one initiator was selected for the artificial aging seeds of Dictamnus dasycarpus Turcz.and one initiator was selected for the artificial aging seeds of Paeonia lactiflora Pall.The viability of the seeds aged 610 days of Dictamnus dasycarpus Turcz.could increase from18.6756.00%to 53.3384.00%after 0.04%ASA initiation.The viability of the seeds aged 1463 days of Paeonia lactiflora Pall.could increase from 33.3386.67%to 44.4495.56%after 0.30%CaCl2initiation. |
