| In order to deeply understand the tea drinking custom of Dracocephalum rupestre Hance in Xinzhou area of Shanxi Province,this study selected villagers from Chunjingwa Village of Ningwu County,Damogou Village of Shenchi County,Jiubao Village of Wuzhai County and Dongyao Village of Jingle County to investigate the difference of the villagers’ understanding in Dracocephalum rupestre Hance,picking habits,tea making process and drinking habits,etc.And it also analyzed the reasons that affect the differences among the four villages.The methods including field investigation,questionnaire survey and interview face to face.The results showed that there were age differences in the identification of Dracocephalum rupestre Hance among the villagers in four villages of Xinzhou,and the proportion of villagers over 50 years old was the highest,the proportion of villagers from 30 to 50 years old was the second,and the propotion of villagers under 30 years old was the lowest.There was no difference between men and women in this respect.The difference of age and sex was also reflected in the cognition of Dracocephalum rupestre Hance blood pressure lowering function.The proportion of villagers aged 30 years and 50 years old was the highest,the second was over 50 years old,and the lowest was under 30 years old,and the proportion of males was generally higher than that of females.There are differences in age and sex in whether or not the villagers drink overnight tea in four villages.The proportion of villagers aged 30-50 years is the highest,the second is over 50 years old,and the lowest is under 30-year-old.The proportion of women is generally higher than that of men,but the proportion of villagers who drink overnight tea is low.When it comes to tea-dringking time,villagers generally choose to drink tea at any time.There were differences in age and sex in drinking tea after meal.The proportion of villagers between 30 and 50 years old was the highest,the second was over 50 years old,and the lowest was under 30 years old.The proportion of men drinking tea after dinner was higher than that of females in Chunjingwa and Damogou villages,while in the other two villages,it is the opposite.The tea-making process of Dracocephalum rupestre Hance varies from village to village.In order to obtain the tissue culture seedlings,accomplish the rapid propagation,and complete the industrialized seedling raising of Dracocephalum rupestre Hance,the effects of disinfection time,hormone ratio,sucrose concentration and light condition on callus induction were studied by inducing the callus of the seeds,leaves and stems of Dracocephalum rupestre Hance.The results showed that when the light condition was 4000 lx,the best explants for callus induction were selected from stem segments of Dracocephalum rupestre Hance;When the light condition was 15000 lx,the leaf was easy to induce callus,while the stem segment was not easy to induce callus,but the induction rate was low.And the optimal disinfection time for mercury was 7 min.The optimum culture conditions were MS + NAA0.5mg/L + 6-BA0.5mg/L + sucrose 30g/L + Agar 7.0g/L;The seeds germinated slowly in culture medium,germinated quickly in petri dish and seedlings were easy to produce callus after root removal.However,the callus size was small,and it was not easy to expand.The best disinfection time was 3 min.The callus of stem segment is easy to take root but not easy to differentiate into bud,but the leaf callus can differentiate bud,but the differentiation rate is low.This study provides a reference for callus induction and plant regeneration of Dracocephalum rupestre Hance. |
