In order to explore the effect of the acidic environment of gastric juice on the absorption and utilization of ferrous glycine sulphate,in this study,based on the research and synthesis of ferrous glycine sulphate,the ferrous glycine sulphate was enteric coated to prepare enteric-soluble ferrous glycine pellets.SD rats were used to explore their iron supplementation and antioxidant capacity.Hydrothermal synthesis of ferrous glycinate.In this experiment,glycine(concentration:3.5 mol/L)and iron powder(glycine and iron powder coordination ratio of 2:1)were used as raw materials,and a stabilizer(citric acid)was added thereto,followed by reaction at 120°C for 12 hours to obtain glycine.Iron(ferrous iron content 17.8%).Through single factor and orthogonal test analysis,the order of conditions affecting ferrous content is: reaction temperature > glycine concentration > reaction time >coordination ratio.The product obtained was confirmed to be ferrous glycinate by infrared spectroscopy and thermogravimetric analysis.Preparation and characterization of enteric glycinate ferrous iron.Preparation of enteric ferrous glycine pellets with polyacrylic resin as enteric coating material,and using response surface methodology and orthogonal experiment to explore the optimal formulation and process parameters of enteric coating liquid,using FTIR,FIB-SEM and intelligent dissolution The instrument is characterized.The results showed that when the mass ratio of the raw material of the coating liquid was 1:1,the concentration of the coating liquid was 0.0592 g/m L,and the concentration of the anti-adhesive agent was 0.0313 g/m L,the ferrous iron of enteric solution could reach95.5% in the simulated intestinal liquid environment.In addition,when the conditions of the coating machine are: the fan frequency is 17 Hz,the liquid supply speed is 8r/min,and the material temperature is 43°C,the enteric ferrous glycine particles reach a complete rate of 96.7%.According to FTIR and FIB-SEM analysis,the structure of ferrous glycine sulphate in the pellet was complete,the thickness of the drug-coated coat in the pellet was 187.8 nm,the thickness of the coat was 38.8 nm,and the thickness of the enteric coat was 668.6 nm.Verification of iron supplementation and antioxidant effects of enteric ferrous glycine.70 iron-deficient SD rats(iron-deficient feeding for 2 weeks)were randomly divided into 7 groups and fed with iron deficiency,ferrous sulfate,ferrous fumarate,and enteric ferrous glycine(1:1).ferrous glycine(1:1),enteric ferrous glycine(2:1)and ferrous glycine(2:1)feeds were analyzed after 4 weeks.The results showed that the growth performance of the rats in the iron supplement group was not significant.The difference was better than that of the iron deficiency group(P < 0.05).There was no significant difference in liver and kidney iron content between the iron supplement groups(P>0.05),while the iron content in the spleen of the mice with enteric ferrous glycine(1:1)and enteric ferrous glycine(2:1)was significantly higher.In the other groups(P<0.05).In the analysis of serum biochemical indicators,the results of T-SOD and MDA showed that the antioxidant capacity of enteric ferrous glycine(1:1)was significantly better than that of ferrous glycine(1:1)and other iron supplements.The index of T-SOD in(2:1)of enteric ferrous glycine(2:1)was significantly higher than that of ferrous glycine(2:1)(P<0.05),but the difference of MDA between the two was not significant.According to the slope ratio method,the relative bioavailability of each iron supplement relative to ferrous sulfate was determined.The results of iron addition of ferrous glycine(1:1)and ferrous glycine(2:1)were better.And the bioavailability of the ferrous iron is further improved after coating.The results of WB and q RT-PCR showed that the iron supplementation effect of ferrous glycine(1:1)was significantly increased after coating(P<0.05),but the result of enteric ferrous glycine(2:1)was not satisfactory.According to the existing results,the iron supplementation and anti-oxidation ability of ferrous glycine(1:1)and ferrous glycine(2:1)and their respective enteric coatings are superior to ferrous sulfate and ferrous fumarate.The ferrous glycine(1:1)and the ferrous glycine(1:1)are coated with an enteric coating to improve the iron supplementation effect.
