As the recommended high-toxic pesticide subsititute,Deltamethrin has a wide range of applications in killing sanitary pests and treating ectoparasites diseases,which may affect the growth and the quality of poultry products.Therefore,it is of great significance to study the enrichment and elimination of deltamethrin in laying hens and toxic effects.The research is mainly divided into the following three parts:Optimization of analytical method of deltamethrin residues.To choose 0.6%,0.4%,0.4%and 6%acidified acetonitrile were respectively used as extraction of tissues,eggs,plasma and manure,which were purified by solid phase extraction and analyzed by gas chromatography-mass spectrometry with selective ion monitoring mode.The results showed that the linear relationship was good in the range of5.0~1000.0?g/L(R~2>0.99).LOD are 5?g/kg,LOQ are 15?g/kg;The recovery rates were more than 81.5%.The intra-day coefficient of variation was less than 15.85%,and the inter-day coefficient of variation was less than 17.86%.The method is accurate and reliable,suitable for the determination of deltamethrin residues in laying hens.Study on the enrichment and elimination of deltamethrin in laying hens.A total112 52-week-old Nongda No.3 laying hens were selected and randomized into 14groups with 8 per groups.One group was basal without deltamethrin.The others were test groups.Test is divided into administration period(14d)and withdrawal period(21d),The dose was 20mg/kg·BW·d,The tissues,eggs,plasma and manure were collected after administration 1 d,3 d,7 d,10 d,14 d and withdrawal 1 d,2 d,3 d,7 d,10 d,14 d,17 d,21 d and determined deltamethrin residues,which obtain the enrichment and elimination in the laying hens.The results showed that the residual concentration of deltamethrin was manure>lung>fat>pouch>egg>plasma>spleen>glandular stomach>heart>kidney>muscle>liver>crown>muscular stomach after administration 14 d,and the residual concentration was fat>egg>manure>plasma>lung>kidney>heart>liver after withdrawal 21 d.The elimination rate of deltamethrin is from fast to slow:liver>lung>kidney>manure>heart>plasma>fat>eggs.Study on toxicity test of deltamethrin on laying hens.1)Deltamethrin had no significant effect on the relative weight of organ of laying hens(P>0.05).2)The activity of glutamate aminotransferase and cholinesterase decreased gradually with the increase of deltamethrin administration time(P<0.05).Plasma urea nitrogen,uric acid,cholesterol,triglyceride and creatinine were increased significantly during the administration period(P<0.05).3)The deltamethrin residues caused obvious liver lesions and loss of intestinal villous.Intestinal wall thickness,villus height and crypt depth were increased,but villus height/crypt depth were opposite.The intestinal self-repair ability is better than liver after withdrawal.In conclusion,deltamethrin residues have extremely adverse effects on the safety of poultry products.Therefore,to choose deltamethrin should pay attention for dosage and interval in cultivation.It is great significance for ensuring poultry products safety and protecting human health and safety.
