| The rice field eel,Monopterus albus is not only delicate in meat,rich in nutrients,high in protein content and very low in fat and cholesterol,but also has extremely high medicinal value,which is an important aquaculture animal in China.At present,the seedlings required for aquaculture are mainly dependent on wild seed,and as the wild resources are reduced,many geographically isolated places of Monopterus albus begin to circulate.It is imperative to investigate the genetic structure of Monopterus albus and protect its genetic diversity.However,at present,the breeding of Monopterus albus is mainly based on introduction and selection,the process of genetic improvement is slow,and the varieties are lacking.Therefore,it is particularly important to improve the breeding efficiency through early identification and accurate selection of target traits in the early stage of breeding.In this study,eight genetic traits of Astragalus membranaceus with obvious traits were selected from Henan,Anhui,Hunan and Hubei province.The hybrid breeding experiment between Monopterus albus in Henan and Chaohu was selected to provide a theoretical basis for breeding good varieties.The specific results are as follows:(1)In this study,the systematic design of the experimental scheme was carried out by bioinformatics analysis.223 wild scutellariae were selected as experimental materials,the rice genome was used as a reference,the SLAF label was obtained by high-throughput sequencing,and finally a large number of specificities were developed on the polymorphic SLAF label.SNP locus.The results showed that 430,983 SLAF tags were obtained in the experiment,among which 193,378 were polymorphic SLAF tags.Through sequence analysis,1,992,507 SNPs were developed,and 93,112 highly consistent population SNPs were screened.Then,using mathematical statistics method,8 jaundice individuals completed phylogenetic tree,population structure,PCA analysis,and found that the scutellaria in these 8 locations are from four different ancestors,due to the geographical location,the jaundice in these 8 locations is Genetic differentiation occurs during growth and development.Through the phylogenetic tree,it is found that the scutellariae distributed in Anhui has a close relationship,and the scutellariae distributed in Songzi,Public Security and Changhu have close kinship.(2)The experiment was carried out in Anhui Chaohu self-crossing seedlings,Henan Gushi self-crossing seedlings,Anhui Chaohu and Henan Gushi hybrid seedlings,and Jingzhou self-crossing seedlings in Hubei,using FRP small cages for breeding,after8 weeks of feeding.The growth indicators of four species of Astragalus membranaceuswere analyzed.The results showed that the growth rate of F1 seedlings between Gushi and Chaohu was the fastest,and the body length increased from 4.23 cm to 12.66 cm on June 8th and the body weight reached 1.9780 g.In the self-crossing population,Gushi self-crossing F1 generation Mully growth and body weight gain reached the maximum,7.31 cm and 8.4607 g,respectively,while the survival rate was the lowest,only 68%. |
