1.This study was designed to observe and analyze developmental dynamics of two growth-related traits(body weight(BW)and standard length(SL))in the three populations of Monopterus albus(population A means deep yellow and big spots,population B means pale yellow and thin spots and C means steel grey).The results shown that the highest absolute growth rate of body length and weight were observed in population A(0.08mm/d,0.25g/d),the second was population B(0.05mm/d,0.16g/d)and the third was population C(0.02mm/d,0.12g/d).There was significant difference of growth speed in population A compared with that in B and C.2.To analyze the nutritive composition in the muscle of three populations of Monopterus albus.Moisture,crude protein,fat,ash,amino acids,fatty acids and mineral elements were investigated by national standard method.The moisture,crude protein,fat and ash consisted of 74.77%-77.20%,17.84%-18.52%,2.51%-4.32%,0.96%-1.01%,respectively.There was significant difference of moisture and crude protein and crude fat in C compared with those in other two populations(P<0.05).Ash in muscle of C was significantly higher than A(P<0.05),but for C and B,it had no significantly difference(P>0.05).There was significant difference of crude protein in A and B(P<0.05).The total content of amino acids was 16.68-17.99g/100g,it had significant difference in C compared with that in other two populations(P<0.05),but it had no significant difference in A and B(P>0.05).The content of essential amino which was 7.16-7.25g/100g from three populations had no significant difference(P>0.05),the content of essential amino acids in A and B were higher than that in C.21 kinds of fatty acids were determinated,the total content of eicosapntemacnioc acid and docosahexaenoic acid which in A and B were higher than those in C were high,about 9.47%-9.99%.The content of mineral elements in muscle was abundant,there were significant difference of 10 kinds of elements in C compared with those in other two populations(P<0.05),only 2 kinds of elements had significant difference in A compared with those in B(P<0.05).It can be concluded that the nutritive composition in muscle of the two populations with yellow skin had no obviously difference,however,they had significantly difference with C population.3.Sequence related amplified polymorphism(SRAP)markers were used to analyze the genetic diversity among three populations(A,B and C)of Monopterus albus.Total of 172 loci were amplified using 10 specific and stable primers selected from 88 primers.The percent of polymorphic loci of three populations were 69.29%,73.23%and 74.04%.Nei'S gene diversit were 0.2781,0.3011 and 0.3126.Shannon'S information index were 0.4065,0.4373,0.4505,respectively.The Nei'S unbiased genetic distance and genetic identity among the three populations was from 0.0410 to 0.0887 and 0.9151 to 0.9598.UPGMA cluster analysis showed that there was closer genetic relationship between A and B with the least genetic distance(0.0410),and farther genetic relationship between the A and C with the most genetic distance(0.0887).4.We had taken 64 pairs of SRAP primers to screen the body colour special band from three populations(A,B and C)of Monopterus albus.At last,we had acquired a special band with Me8/Em8 primer combination.Cloning and sequencing the special bands,four pairs of 20bp primers were designed acorrding to the sequence of band,amplified the three populations' genomic to get the same size goal band.The results showed the SRAP mark didn't convert to simple SCAP markers.
