This experiment was carried out in the Laboratory of Small Berry Germplasm Resources and black currant Germplasm Resources of Northeast Agricultural University from 2017 to 2019.The main varieties of black currant have a bad phenomenon in the actual production-bud second sprout,resulting in imbalance of tree potential,loss of nutrients in the tree,weakening of stress resistance,seriously affecting the development of the industry,urgently to be resolved.On the basis of the previous research of this group,this experiment identified the second germination by high performance liquid chromatography with the easy bud second sprout variety “Adelinia” and the non-second germination variety “Brodtrop” “Hei Feng” as the test material.Species of phenolic substances and changes in content during bud second sprout,combined with phenylalanine ammonia-lyase?PAL?,chalcone isomerase?CHI?,polyphenol oxidase?PPO?,cinnamic acid-4-Changes in key enzyme activities such as hydroxylase?C4H?,determine the metabolic characteristics of phenolic substances,and regulate bud second sprout using exogenous 30 mg/L GA3 and 50 mg/L ABA for all identified phenolic substances and The key enzyme activities were analyzed to further verify the relationship between phenolic substances and bud second sprout,which laid a theoretical foundation for the final re-emergence of black currants,and provided data support for the artificial regulation of black currants' bud second sprout.Development provides scientific security.The main findings are as follows:?1?There are mainly 8 main phenolic substances in the second sprouting of black currant,which are Quercetin,Myricetin,Epicatechin,Isorhamnetin,Gallic acid,Kaempferol and Dihydroanthracene.Peel and Ferulic acid;Quercetin,Myricetin and Epicatechin a re found in the germination buds of the three varieties,which are the main phenolic substances and the least Gallic acid content.The types and contents of different types of phenolic substances are also different.It is not easy for the bud second sprout varieties “Brodtrop” and “Haifeng” to contain the highest content of Quercetin,which is 90.72 mg/g and 101.36 mg/g,which is 1.06,1.18 times of "Adelinia";easy to bud second sprout variety "Adelinia" contains the highest content of Myricetin,reaching 93.21 mg / g,which is 1.31,1.37 times of the others.?2?During the second germination process of black currant,the content of different phenolic substances is different,and the effect on germination is also different.The contents of Ferulic acid,Epicatechin and Gallic acid decreased gradually with germination,which may be the inhibitor ofgermination;The content of Myricetin,Kaempferol and Dihydroquercetin gradually increased with germination,which may be germination.The quercetin content fluctuat ed with the germination,and the final content increased slightly;The content of Isorhamnetin fluctuated with the germination,and the final content was basically unchanged;the two may have little effect on germination.During the bud second sprout process of black currant,the Ferulic acid,Epicatechin and Gallic acid content of the bud second sprout varieties “Brodtrop” and “Heifeng” were always higher than the easy bud second sprout variety “Adelinia”;The content of Myricetin in the bud second sprout variety “Adelinia” is always higher than that of the bud second sprout varieties “Brodtrop” and “Heifeng”.?3?The total phenolic content of black currant during germination has an inhibitory effect on germination.With the germination,the total phenolic content in the second germination of black currant decreased rapidly,and the easy-to-second germination variety “Adelinia” decreased more than the bud second sprout varieties “Browde” and “Hei Feng”.It is 1.50 and 1.59 times of both.?4?The key metabolic enzymes PAL,CHI,C4 H in the second germination process of black currant have a certain inhibitory effect on germination.With the germination,the activities of PAL,CHI and C4 H in the second germination of black currant decreased rapidly,and the PAL activity of the second germinated variety “Adelinia” decreased more than that of the second germinated variety “Brodtrop”."Hei Feng",and the decline in the activity of CHI and C4 H of the three varieties is not much different.The key enzyme of PPO,a key enzyme in the process of bud second sprout of black currant,has a certain promoting effect on germination.With the germination,the PPO activity in the second germination of black currant increased rapidly,and the PPO activity of the second germination variety “Adelinia” decreased less than that of the non-second germination varieties “Brodtrop” and “Hei Feng”.?5?Exogenous GA3,ABA can regulate the total phenolic content in the second sprout and regulate the bud second sprout process.After exogenous application of GA3,the total phenolic content in the buds decreased,which promoted the bud second sprout of bla ck currant.ABA can inhibit the bud second sprout,and the total phenol content in the bud is inhibited and maintained at a high level.?6?Exogenous GA3,ABA can regulate the key enzyme activities of phenolic metabolism in the second sprouting and then regulate the bud second sprout process.During the bud second sprout of black currant,GA3 treatment promoted the decrease of PAL activity,ABA inhibited the decrease of PAL activity,GA3 treatment promoted the increase of PPO activity,ABA inhibited the increase of PPO activity,and GA3 treatment promoted CHI activity.The decline of ABA inhibited the decrease of CHI activity;GA3 treatment had little effect on the change of C4 H activity,and ABA inhibited the decrease of C4 H activity.?7?Under the same treatment,the content of phenolic substances iin the second sprout of black currant and the changes of key enzyme activities of phenol metabolism may vary from species to species.Exogenous ABA treatment had significant effects on the ch anges of totalphenolic content and PPO activity in the second sprout varieties;the regulation of exogenous GA 3 on total phenolic content,PPO activity and C4 H activity of the second sprout varieties The effect is remarkable,and the effect of regulating the change of PAL activity of the second sprout varieties is remarkable.
