Chicken colibacillosis is the most common and frequently-occurring disease in poultry farming.Environmental factors,stress response or secondary disease can lead to the onset of disease.Clinically,antibiotics are used for treatment,resulting in the emergence of drug-resistant strains.The curative effect is not as good as before,and it has a bad influence on human health and living environment.Numerous clinical practices and modern Chinese medicine pharmacology studies have confirmed that many traditional Chinese medicines have played an active role in the treatment of bacterial infections.However,there are many kinds of veterinary drugs in the market,and some prescriptions for veterinary drug preparations lack the theoretical basis of Chinese veterinary syndrome differentiation.Therefore,this experiment uses different models of E.coli infection to observe the characteristics of symptoms and signs and the characteristics of Chinese veterinary syndrome,establish the syndrome differentiation standard of chicken colibacillosis,and provide theoretical basis for the development and clinical promotion of Chinese veterinary drugs for prevention and treatment of chicken colibacillosis.Test 1:Establishment and observation of a model of chicken Escherichia coli inoculated with a laryngeal tracheal tube.The pre-test showed that the dose of the tracheal tracheal inoculation was 0.54×109 CFU/mL bacterial solution 0.5 mL/only,the incidence rate could reach 100%,and the mortality rate was 40%.Symptomatic lesions are basically consistent with clinical natural onset,so the model was established by injecting this dose through the throat tracheal inoculation.after toxicity attack,observe the clinical symptoms and changes in body temperature,and at the same time,the blood biochemical,histopathological sections and inflammatory factors related indicators were detected.Results:Symptoms and tissue and organopathy are constantly changing with the development of the disease.At 12 h after toxicity attack,the main symptoms were febrile fever,accounting for 100%of the total number of surviving flocks.At 36 h after toxicity attack,90%of the surviving chickens showed typical diarrhea symptoms.At 72 h after toxicity attack,the chicken population temperature began to appear.Decline;the necropsy found exudative pericarditis in the dead chicken 24 h after toxicity attack;48 h after toxicity attack,the liver showed lesions,marginal congestion and necrosis,the detection rate of typical lesions such as pericarditis and perihepatitis after 96 h after toxicity attack Up to 100%,individual chickens have kidney,spleen enlargement and other lesions;pathological observation found that 12 h after toxicity attack,the lungs first appeared lesions,and then invaded the heart,intestine,liver and other tissues and organs;indicating that bacteria invaded the body after the respiratory tract through the lungs into the blood,and gradually invade other tissues and organs of the body.Blood biochemical indicators showed that ALT,AST and TBIL increased significantly and significantly increased at 24 h-36 h(p<0.05,p<0.01),and AST decreased significantly at 72 h after toxicity attack p>0.05).The detection of serum index showed that the level of IL-1β in serum increased at 24 h after toxicity attack,and was significantly different at 24 h,36 h,48 h and 72 h(P<0.05),and returned to normal at 72 h after toxicity attack.The TNF-a content was in the early stage of challenge.There was no significant change,and there was a significant increase at 96 h after toxicity attack(P<0.05),indicating that the inflammatory response in the development of the disease is increasing and the liver function is seriously impaired(P<0.05).The above results indicate that the artificial disease model can be successfully established by injecting chicken E.coli through the tracheal trachea and simulating the natural infection pathway,which provides a reference for the artificial establishment of the chicken colibacillosis model and the screening of traditional Chinese medicine for the treatment of chicken colorectal disease.Test 2:The establishment and observation of the experimental two-injection model showed that the dose of the pectoral muscle injection was 0.30×109 CFU/mL bacterial solution 0.5mL/only,the incidence rate reached 100%,the mortality rate was 50%-70%,symptomatic lesions.It is basically consistent with clinical natural onset.after toxicity attack,the relevant indicators were observed as above,and blood,bacteremia and liver contact related indicators were detected.RESULTS:The time points of the symptoms and lesions in this model were earlier and more severe than the throat tracheal inoculation model.Fever symptoms occurred 12 h after toxicity attack,and fever symptoms disappeared after 24 h.The duration was 12 h lower than that of the laryngeal tracheal inoculation model.Some chicken body temperature began to decrease,36 h earlier than the laryngeal tracheal inoculation model;the diarrhea symptoms were 24 h after toxicity attack.Appeared 12 h earlier than the tracheal tracheal inoculation model,accounting for 85%of the surviving flocks;the time points of pericarditis and perihepatic lesions were more than 12 h earlier than the laryngeal tracheal inoculation model,and at 60 h,The detection rate reached 100%;the time of observation of the pathological changes of heart and liver was also observed 24 hours earlier.Blood routine examination showed that the total number of WBCs in the challenge group was significantly higher than that in the control group at 36 h(p<0.05);it was significantly higher at 60 h,72 h,96 h,120 h,and 144 h than the control group(p<0.01).The number of neutrophils(NEUT)increased at 72 h after toxicity attack,which was significantly higher than that of the control group(p<0.01).There was no significant difference between blood biochemical index and inflammatory factor test and laryngeal tracheal inoculation model.Both of them reflect that the bacteria are intensifying after invading the body.The detection rate of the test chicken bacteremia was as high as 87.5%at 12 h after toxicity attack.The pathogens were observed in the liver tissue of the dead chickens in each time period,indicating that the bacteria could be attacked by intramuscular injection.Rapid entry into the bloodstream,causing bacteremia,significantly reducing clinical infections and morbidity.The above results indicate that the intramuscular injection model has the characteristics of rapid onset,strong pathogenicity,small required dose and high mortality,and lacks a typical course of clinical natural infectionTest 3:The best method for the differentiation of the three chickens with Escherichia coli is the syndrome-differentiation of the six meridians and the syndrome differentiation of Weiqi Yingxue.The disease course of chicken Escherichia coli in the tracheal tracheal inoculation model was analyzed according to the syndrome-differentiation of the six meridians:6 h-18 h after toxicity attack,the diseased chickens mainly showed aversion to cold(crowding,fluffy feathers,etc.)and high fever(increased body temperature of more than 80%of the sick chickens)Above 1℃,the spirit is extremely depressed,etc.),the disease location was transferred from the Taiyang disease to Yangming disease;18 h-24 h after toxicity attack,chickens with fever symptoms accounted for only 33%of the surviving chickens.This is consistent with the fact that the syndrome of Yangming disease is transferred to the syndrome of Shaoyang disease,and the heat is not retreating,and the heat and cold are consistent.After 24 h to 36 h after toxicity attack,the chickens in the diseased chickens are gradually reduced,and the body temperature is gradually decreased.And 90%of the sick chickens have diarrhea symptoms,which is basically consistent with the syndrome of Taiyin disease;36 h-72 h after toxicity attack,This is consistent with the cold,drowsiness and dizziness of Shaoyin syndrome,At this time,the disease is in the heart and kidney;after 72 hours,the compliance degree of cold and heat mixed with Jueying symptoms reached 95%.According to the the syndrome differentiation of Weiqi Yingxue:6 h-18 h after toxicity attack,This is consistent with the main syndrome with the Weifen,the disease was in the Weifen;24 h-48 h after toxicity attack,and the disease into the blood at the stage of the Yingfen.This is consistent with the main syndrome with the Yingfen;after toxicity attack 48 h-72 h,individual chicken,kidney enlargement,blood function analysis,liver function was seriously reduced,and blood disease was mainly caused by dizziness,liver and kidney disease,and the disease was in Xuefen.In the intramuscular injection model group,the course of the chicken was analyzed according to the syndrome-differentiation of the six meridians:6 h-12 h after toxicity attack,This is consistent with the main syndrome with the Taiyang and Yangming,and the disease position was Taiyang to Yangming;12 h-24 h after toxicity attack.At 24 h,febrile chickens accounted for only 8%of the total number of surviving chickens,while the proportion of diarrhea chickens reached 85%.Therefore,the disease status was identified as Shaoyang introduction of Taiyin syndrome;24 h to 72 h after toxicity attack,This is consistent with the main syndrome with the Shaoyin;72 h after toxicity attack,the temperature of the chicken group-showed a significant decrease,dizziness was exhausted and the liver and kidney showed lesions,and the disease was in the Jueyin.According to the syndrome differentiation of Weiqi Yingxue:6 h-12 h after toxicity attack,This is consistent with the main syndrome with the Weifen,the disease was in the Weifen;12 h-24 h after toxicity attack,the disease is into the blood at the stage of the Yingfen.This is consistent with the main syndrome with the Yingfen.24 h to 72 h after toxicity attack,the symptoms decrease with body temperature.Drowsiness-based,accounting for more than 80%of the total number of surviving chickens,typical peritoneal pericarditis and perihepatitis can be seen by necropsy,the detection rate is 100%,individual chickens show spleno-renal enlargement,and blood disease is dizzy Liver and kidney disease,at the stage of the Xuefen.In this study,an artificial model of chicken Escherichia coli was established through two methods,laryngeal tracheal inoculation and intramuscular injection.The two models were compared and analyzed in depth,and dialectically through the syndrome-differentiation of the six meridians and the syndrome differentiation of Weiqi Yingxue.Preliminary establishment artificial disease model and syndrome differentiation criteria for chicken colibacillosis.
