Andrographis herba an aerial dried part of Andrographis paniculata(Burm.f.)Nees in Acanthaceae,which is slightly fragrant and extremely bitter.The main medicinal ingredients of A.paniculata are andrographolide and dehydroandrographolide,which are widely distributed in leaves.Clinically,A.paniculata is used to prevent upper respiratory tract infections,gastrointestinal and hepatobiliary diseases.The commercial medicinal materials of A.paniculata are artificially cultivated varieties.Although its planting area is extensive,due to factors such as planting environment and technology,the varieties of A.paniculata are degraded,the medicinal components are variously and unstable,and the seeds of A.paniculata are miainly provided by the seed fields in Guangxi.The genetics simple,is not conducive to the breeding of A.paniculata.This study analyzed the growth and material accumulation of A.paniculata in Hangzhou,which seeds were space flight with ’The long March 7’ and ’Shenzhou 11’.The effects of aerospace mutagenesis on the biological characters and active constituents of SP1 and SP2.This study provided a theoretical basis for the selection of high quality and high yielding new varieties of A.paniculata.The main results are as follows:1.Study on the growth and material accumulation of Andrographis paniculata.Through the study of agronomic traits of A.paniculata planted in Hangzhou in various periods,we found that the growth trend of A.paniculata showed an "S’ shape,the highest growth rate in the growth period and decreased after budding period.The fresh and dry weight of the plant reach the maximum at the flowering stage,while the leaf weight ratio shows a downward trend with the growth of A.paniculata,reaching a minimum of 25.39%at the fruit stage,which does not conform to the leaves should not be less than 30%in ChP.Through the study on the physiological change rule of A.paniculata in each growth period,we found that the soluble protein content shows a slowly rising trend;The soluble sugar reaches the maximum value in the first rapid growth period,then falls sharply to the minimum value in the second rapid growth period;The value of total flavonoids increased first and then decreased.The HPLC method was used to determine the content of the active constituents of A.paniculata.It was found that the content of andrographolide in leaves and stems reached the maximum at flowering stage and initial flowering stage respectively.The content of deoxyandrographolide reached the maximum at the seedling stage.The content of deoxyandrographolide in the leaves of seedlings reached the maximum,while the content of dehydroandrographolide in stems and leaves change unconspicuous.Based on the comprehensive consideration,we found that the content of andrographolide is the maximum in the full flowering stage,so it is suggested that A.paniculata planted in Hangzhou should be harvested during the flowering perioda.2.Study on SP1 generation of Andrographis paniculata by Space Mutation.The effects of space mutagenesis on SP1 generation of A.paniculata were studied by measuring 10 agronomic traits such as plant height,leaf length and leaf number of SP1 generation of A.paniculata and the content of 4 kinds of diterpene lactones.Studies have found that the traits of SP1 generation of A.paniculata showed different degrees of variation after aerospace mutagenesis treatment.The leaf weight ratio had a very significant positive correlation with the number of primary tillers,leaf dry weight and dry weight,and a very significant negative correlation with the content of deoxyandrographolide.The content of andrographolide was significantly negatively correlated with the number of leaves and the number of primary tillers,and was positively correlated with the other three diterpene lactones.Principal component analysis was carried out on 14 characteristics of A.paniculata,and 5 principal components were extracted,which were yield factor,plant type factor,leaf type factor,component factor and seed weight factor respectively,indicating that yield factor was dominant in the selection and evaluation of A.paniculata..3.Study on SP2 generation of Andrographis paniculata by Space Mutation.The cultivation of A.paniculata SP2 generation with characteristic morphology.We found that the growth trend of CZ-3,CZ-119(trifoliate symmetry)and SZ-85(main branch)were significantly higher than CK,while the growth trend of CZ-88 and CZ-118(main branch)were similar to CK.At the same time,the biomass and active ingredient content of each strain were measured.It was found that the fresh weight and dry weight of CZ-88 strain were significantly higher than CK,and the content of andrographolide reached the maximum in leaves and stems.Based on the dual control of yield factor and quality of SP1 generation,CZ-88 can be regarded as the focus of SP3 generation breeding of A.paniculata.
