Salvia has been used as a traditional medicine in China for thousands of years.Chinese scholars' research on the chemical constituents of Salvia miltiorrhiza began at the end of the last century, and a lot of work has been done to separate and extract the water-soluble components. The active ingredients of Salvia miltiorrhiza include fat-soluble and water-soluble,and then polysaccharides and volatile oils are extracted from Salvia miltiorrhiza. In this thesis, the extract of Salvia miltiorrhiza obtained from the refined salvia miltiorrhiza in Anhui Province was used as the research object.20 g Kunming mice were used as experimental subjects, and the recommended doses were 10 times,5 times and 1 times. The drug concentration of the extract was 0.0195 g/mL,0.039 g/mL,and 0.078 g/mL for 28 days. The serum of mice was taken regularly, and the concentration of various cytokines was detected by ELSIA method. The regulation of the immune activity of Salvia miltiorrhiza extract on mice was studied. After 28 days of intragastric administration,Escherichia coli was injected intraperitoneally. After the necropsy, the intestinal tissues of the mice were examined by pathological section to observe the protective mechanism of Salvia miltiorrhiza extract and intestinal structure.1.The results showed that the appropriate concentration of Salvia miltiorrhiza extract can promote the growth of healthy mice, improve the body's immunity by increasing the index weight of immune organs and increasing the content of ALB and Ig in serum, which is basically consistent with the theoretical prediction.High-dose drugs can stimulate the secretion of inflammatory factors in the body, and there may be potential predictions that trigger the body's inflammatory response.2.E.coli challenge test was carried out on mice pre-administrated with Salvia miltiorrhiza extract.By measuring the concentration of cytokines, biochemical indicators and death in the serum of mice,it was concluded that the extract of Salvia miltiorrhiza was small after attacking E.coli.The results showed that the extract of Salvia miltiorrhiza can significantly reduce the mortality of E.coli, inhibit the pro-inflammatory factors of mice after challenge, increase the serum Ig G concentration, and improve the immunity of mice, and the dose is increasing. And the Salvia miltiorrhiza extract can also protect the body from liver damage caused by LPS released by E.coli.3.The mice after challenged with E.coli were sliced by pathological tissue section and the like, and the intestinal villus structure and mucosal state of the mice were observed. The results of this experiment showed that the pathogenic E.coli O101 had the most significant ileal injury in mice. The villus was basically broken and fractured, and the intestinal wall became thicker. In the early stage, the test mice were pre-administered with different concentrations of Salvia miltiorrhiza extract for 28 days.Toxic and necropsy were used to make intestinal pathological tissue sections. It was found that the intestinal structure of the mice had significant protective effects compared with the model group. The intestinal structure was relatively complete and the villi arrangement was dense. The high-dose group ofSalvia miltiorrhiza extract has a significant effect on the protection of the intestinal structure of the jejunum and ileum caused by E.coli O101.
