At this stage,more than 90% of artificial insemination adopts fresh essence preserved at 17?,but the storage time of pig semen is relatively short at 17?,generally 3 to 7 days,and is susceptible to microbial contamination,which is not conducive to improving excellent boar semen.Utilization rate increases the cost of improving pig breeds.Low temperature of 4? can not only effectively reduce the physiological metabolism of sperm,but also maintain semen quality and prevent microbial contamination for a long time.However,at present,the research on the cryopreservation of pig semen at 4? is still not deep,and further research and practice are needed in the methods of cryopreservation of semen and the active components of diluted preservation solution.This experiment is mainly aimed at the key technical aspects such as semen treatment method and active ingredient of dilute solution in pig semen 4? cryopreservation,and carries out detailed research to provide theoretical and practical support for the wide application of pig semen 4? cryopreservation technology in pig production enterprises.(?)Study on the technical method of 4? preservation of pig semen1.Research on cooling methods.Using a self-made thinner formula,after artificial semen collection,the sperm concentration of 200 million / m L,milky white,normal odor and sperm viability of more than 80% of the pig essence,3 times diluted,the control group used a layer of towels The parcels were wrapped in four layers of towels and four layers of towels + foam boxes.This experiment was repeated 3 times(the same below).The results showed that the sperm motility and vigor were 79.69% and 63.81%,respectively,which were significantly better than the control group(P<0.05).2.Cooling equilibrium time study.On the basis of the above test,the pre-dilution operation was the same,and after four-layer towel + foam box wrap,the step was cooled.First,in the 17? for storage.The results showed that the sperm concentration and vigor were 76.35% and 65.34%,respectively,when the temperature was constant at 17? for 2.5 h,which was significantly better than that of the control group(P<0.05).3.Dilution multiple study.Using the homemade thinner formula,after artificial semen collection,the sperm concentration is about 200 million/m L,and the porcine essence is more than 80%.The dilution ratio of the control group is 1:1,and the dilution ratio of the test group is 1:2,1:3,1:4,and then save using the best cooling balance method described above.The results showed that the sperm motility and vigor were 78.13% and 69.65%,respectively,which were significantly better than the control group(P<0.05).(?)Effects of fructose and sodium alginate on the preservation of pig semen at 4?1.On the basis of the pre-treatment test,the fructose of 0,0.2,1,2,4 mg/m L was added to the BTS base diluent to investigate the effect of different concentrations of fructose on the preservation of pig semen at 4?.The results showed that when adding 0.2mg/m L fructose,the sperm motility,plasma membrane integrity rate,acrosome integrity rate and sperm rapid motor activity were 79.17%,76.53%,80.58% and 50.1%,respectively,which were significantly better than those of the fructose.Control group(P<0.05).2.Add 0,0.2,1,2,4 mg/m L sodium alginate to the BTS base dilution to investigate the effect of different concentrations of sodium alginate on the preservation of pig semen at 4?.The results showed that the sperm motility,plasma membrane integrity,acrosome integrity and sperm motility were 85.55%,86.33%,87.51% and 70.9%,respectively,when added with 1mg/m L sodium alginate,which was significantly better than 85.55%,86.33%,87.51% and 70.9%,respectively.Control group(P<0.05).(?)The effect of the optimal combination of fructose + sodium alginate on the preservation of pig semen at 4?On the basis of the second step of the experiment,0.2,1 mg/m L fructose and 1,2 mg/m L sodium alginate were used to optimize the combination of diluent components,and the thinner A brand of mature application was used as a control.The effect of fructose + sodium alginate on the preservation of pig semen at 4?.The results showed that: 0.2mg/m L fructose + 2mg/m L sodium alginate was added,and the sperm motility,plasma membrane integrity rate and acrosome integrity rate were 72.85%,71.19% and 79.68%,respectively,on the 7th day,which was significantly better than the control group(P < 0.05).In summary,fructose and sodium alginate can significantly increase the storage time and sperm motility of pig semen at 4?,and the optimum concentrations are 0.2 mg/m L and 1 mg/m L,respectively;after the optimal combination of the two active ingredients,the dilution When fructose 0.2mg/m L and sodium alginate 2mg/m L were added to the solution,the semi-preservation effect of pig semen at 4? was the best,and the sperm motility,plasma membrane integrity rate,acrosome integrity rate and Sperm fast movement vitality.
