| With the development of broiler industry to scale and industrialization,whether in artificial insemination or natural breeding,rooster semen quality is the key to affect the reproductive capacity of broilers.Proper dilution of spermatozoa and pretreatment under certain conditions are beneficial to improve the quality of semen at room temperature and low temperature,and to make full use of the good breeding male.In this study,AA white feathered broilers were selected as samples,and a single factor completely randomized experimental design was adopted to compare the effects of semen preserved with different diluents at different temperatures and under the conditions of semen removal and purification on the semen quality of white feathered broilers.The results provided technical support for the breeding of white feathered broilerS。The main contents and results are as follows:1.Different diluents and different time points at room temperature storage conditionsThree kinds of semen diluents were prepared(self-mixing liquid A,self-mixing liquid B and BPSE),and the three diluents were in the proportion of 1:2 to the original semen solution.With the increase of preservation time,sperm motility of liquid B was significantly higher than that of liquid A and C after 2h preservation(P<0.01),but there was no significant change between liquid A and C(P>0.05).The effective sperm count of liquid B was significantly higher than that of liquid A(P<0.01)and that of liquid C(P<0.05)after being placed at room temperature(37℃)for 2h.In general,sperm motility,viability and effective sperm count of liquid B were basically maintained at a higher level than those of the other two groups.2.Different diluents and different time points at low temperature storage conditionsPrepare three diluents of semen(liquid B,liquid D,and E).The three diluents are prepared according to the ratio of 1:2,and stored at a low temperature of 4℃ for 0,2,4,8,24h,and measured Semen quality.After 8h,the sperm motility of E fluid was significantly higher than that of B fluid,and the sperm vitality of E fluid was significantly higher than that of D fluid.In general,sperm motility was maintained at a higher level,and fluid E motility was higher than that of the other two groups.3.Effect of removal of seminal plasma preservation on semen qualityMixed semen was equally divided into 4 groups(2 seminal fluid group and 2 seminal plasma group)and diluted at different proportions and kept at low temperature for 192h.During this period,semen quality was checked every 24h. |
