| Stress has an adverse effect on the growth and reproductive performance of animals.The cage stress inhibits the breeding of goose breeding.Gonadotropin inhibiting hormone(GnIH)can inhibit the breeding of goose by acting on gonadotropin releasing hormone(GnRH)and pituitary gland to reduce the synthesis and secretion of gonadotropin.The effect of cage stress was similar to that of GnIH,suggesting that GnIH may have a synergistic effect in the reproductive performance of cage-inhibited goose.In the first experiment,the male goose was raised in two modes: cage culture and flattening.The differences in reproductive performance between the two types of male geese were compared.The mechanism of GnIH in the reproductive performance of cage-inhibited male goose was studied.The results showed that compared with the Pingyang group,the weight of the liver(P ? 0.05)and spleen(P ? 0.05)in the cage group decreased,and the liver and spleen tissues were damaged to varying degrees,and the testicular volume became smaller(P ? 0.05).Cage globulin(GLB)was significantly elevated(P < 0.05);albumin(ALB),superoxide dismutase(SOD),triglyceride(TG)and alkaline phosphatase(AKP)were significantly reduced(P < 0.05).Serum hormone levels were detected in the cage group,adrenocorticotropic hormone(ACTH),testosterone(T),follicle-stimulating hormone(FSH),luteinizing hormone(LH)Both activin and ACT levels increased(P < 0.05).Fluorescence quantitative detection revealed a significant increase in testicular GnIH expression in the cage group(P ? 0.05).The results showed that cage culture can inhibit the reproductive performance of male geese,and it is possible to inhibit the reproduction of male geese by GnIH.In the second experiment,the in vitro culture method of Leydig cells was established,and GnIH was used to express its hormone secretion and gene expression by overexpressing GnIH in stromal cells(GnIH overexpression plasmid and different concentrations of exogenous GnIH,respectively).Impact.The results showed that both GnIH overexpression plasmid and exogenous GnIH significantly reduced the secretion of T(P < 0.01)and increased the secretion of progesterone(P4),ACT and INH(P < 0.01),and consistency is high.Compared with the empty vector,the expression of GnIH and its receptor in the GnIH overexpression group was down-regulated(P < 0.01),and 3β-hydroxysteroid dehydrogenase(3β-HSD)and CYP17A1 were also significantly down-regulated(P < 0.05);cytochrome P450 cholesterol side chain lyase(CYP11A1,CYP19A1),activin receptor IIα(ActR IIα),activin receptor IIα(ActR IIβ),inhibin βA The levels of(inhibin βA,INH βA)and inhibin βB(inhibin βB,INH βB)were elevated(P < 0.01).GnIH at 200 ng/mL significantly down-regulated the expression of GnIH and its receptor(P < 0.01).The results showed that the effects of exogenous GnIH and GnIH on the interstitial cells were different,and the specific mechanism needs further study.This study showed that cage culture can inhibit the reproductive performance of male goose,and GnIH can mediate stress to affect the reproductive performance of male goose;GnIH overexpression and exogenous GnIH can significantly inhibit the secretion of testosterone in interstitial cells. |
