Resistance to powdery mildew is an important trait of interest in many wheat breeding programs.Wheat cultivars DH51302 and Shimai 26 are highly resistant to powdery mildew at the seedling stage,and the sources and information on their resistance to powdery mildew have not been reported.DH51302 and Shimai 26 were crossed to a powdery mildew susceptible cultivar Zhongzuo 9504.Using the genetic linkage maps,the infection patterns against,the analysis of gene-linked molecular markers and the 660K SNP array to understand the F2:3 families were used in molecular localization of the resistance genes,aiming to clarify the source of powdery mildew resistance genes in DH51302 and Shimai 26,to understand the relationship between target genes,and to avoid the reuse of bands in breeding.Parents with the same disease resistance gene provide a scientific basis for parent selection and collocation of wheat resistance to powdery mildew breeding.Fourteen polymorphic markers,which were linked to Pm52 from Liangxing 99,were used to establish the genetic linkage maps for the resistance genes PmDH51302and PmSM26 in DH51302 and Shimai 26,respectively.Genetic linkage maps display,These genes were placed in the same genetic interval where Pm52 resides.The infection patterns against display,winter wheat breeding line DH51302 derived from Liangxing99 and cultivar Shimai 26 derived from Jimai 22 showed identical infection patterns against thirteen isolates of Blumeria graminis f.sp.tritici?Bgt?that causes wheat powdery mildew.It is speculated that the powdery mildew resistance gene in two varieties is an identical gene.Using 660K SNP array to analyze the difference in 2B chromosome composition between Liangxing 99?the parent of DH51302?and Jimai 22?the parent of Shimai 26?,the results showed that the difference between the 2B chromosomes of Liangxing 99 and Jimai 22 was small.Analysis of gene-linked molecular markers indicated that PmDH51302 and PmSM26 differed from other powdery mildew resistance genes on chromosome 2BL,such as Pm6,Pm33,Pm51,MlZec1,MlAB10,and Pm64.The above results indicate DH51302 and Shimai 26carried the same gene,Pm52,which confers their resistance to powdery mildew.
