Cinnamomum camphora L.is also called the Camphor tree,subtropical evergreen broad-leaved forest trees,is not only an important afforestation tree species in south China,it is important timber tree species,the dense trees,evergreen,branches and leaves guangzhan,hard wood,the timber,gardens,chemical and other fields have a wide range of applications,has a high ecological and economic value.In order to optimize and perfect the Camphor tree regeneration system,provide reference basis for genetic transformation,to meet the needs of the Camphor tree material,this study with Camphor tree immature zygotic embryos as explants,embryo in vitro culture technique,study on factors of early germination of immature embryo,different provenance Camphor tree regeneration system of hunan province to build three aspects of research.The main contents and results of the study are as follows:(1)Carry out in vitro culture technology of Camphor tree seeds.Light culture was the main culture method of immature zygotic embryo of Camphor trees.The optimal medium for the germination of Camphor tree immature embryos was MS+1mg.L-1 6-BA+0.1mg.L-1 2,4-D.The optimal medium for secondary proliferation was MS+1.5mg.L-16-BA +0.5mg.L-1 IAA.The optimum rooting medium was MS+1mg.L-1 IBA+O.3mg.L-1 NAA,and the rooting rate was 76.7%.(2)The factors affecting early embryonic development were discussed systematically.The germination rate and germination time of germination could be improved obviously by disinfection of seed with pulp.There was no significant difference in the germination induction of young Camphor embryo between pretreated seeds at low temperature and soaked seeds.The germination rate and average germination number were significantly affected by different seed selection time.The germination induction effect of immature embryos in July was the best,and the immature zygotic embryos in July were the best.The germination rate of embryos in different media ranged from 50%to 90%,and MS was the best medium for Camphor somatic embryogenesis.The germination rate of germination was significantly affected by the genotype of the mother plant.With 150mg.L-1 activated carbon added,the Browning rate of the medium was low.The inositol concentration had no significant effect on the early induction of Camphor immature embryo.Orthogonal design tests were conducted on 6-BA,NAA,IBA and streptomycin,and the order of influence was:6-BA>NAA>streptomycin>IBA.(3)Establish the somatic embryo regeneration system of Camphor trees of different provenance in hunan province.The optimal medium for the induction of somatic embryogenesis in yueyang and huaihua Camphor trees was MS+1 mg.L-1 6-BA +0.1 mg.L-1 2,4-D,liling,changsha was MS+1 mg.L-1 6-BA+0.3 mg.L-1 2,4-D,chenzhou,loudi was MS+1 mg.L-1 6-BA +0.1 mg.L-1 NAA.In the bud induction experiment,5 mg.L-16-BA +0.1 mg.L-1 NAA hormone combination was suitable for bud differentiation of Camphor trees from yueyang,huaihua and loudi,3 mg.L-1 6-BA +0.3 mg.L-1 NAA from liling and changsha,and 5 mg.L-1 6-BA +0.3 mg.L-1 NAA.The embryogenic callus induction rate of Camphor trees in changsha and yueyang the growth was good,followed by Camphor trees in chenzhou and huaihua,and then in tiling and loudi.The maximum induction rates of somatic embryo were 56.41%,42.31%,48.96%,64.71%,63.25%and 56.31%,respectively.The rooting rate of camphor was different in different provenance.The optimum concentrations of hormones for rooting of Camphor buds in yueyang,chenzhou,liling,changsha,huaihua and loudi were 1 mg.L-1 IBA+0.1 mg.L-1 NAA,1 mg.L-1 IBA+0.5 mg.L-1 NAA,1 mg.L-1 IBA+0.3 mg.L-1 NAA,1 mg.L-1 IBA+0.3 mg NAA.
