The content of protein,fat,lactose and minerals in buffalo milk is higher than that in Holstein milk.Buffalo milk is rich in fatty acids,including polysaturated fatty acids and unsaturated fatty acids.Stearyl coenzyme A desaturase 1(SCD1)is an endoplasmic reticulum enzyme that catalyzes the conversion of saturated fatty acids from DE novo synthesis or dietary intake into monounsaturated fatty acids to improve the unsaturated index of fatty acids.In order to fully understand the difference of fatty acid species and content between Italian buffalo and hybrid buffalo milk of Holstein cows,the differences in the expression of fatty acid metabolism-related genes such as SCD1 in the milk of buffalo and Holstein cows and the physiological role of the fatty acid metabolism-related gene SCD1 in the breast epithelial cells of buffalo were explored.This study application of gas chromatography were measured in lactation mid 30 Italian water milk samples 30 hybrid water milk samples and 30 Holstein cows milk samples of types and content of fatty acids and fatty acid composition table is analyzed on the fatty acid profile,and milk composition analyzer is used to inspect the three basic composition differences in milk.RNAi and lentivirus overexpression were used to verify the effect of SCD1 gene on related genes in the process of milk fatty acid synthesis.The results show that:1.In terms of the amount of fatty acids per 100 g of cow's milk,the contents of various fatty acids in buffalo and hybrid buffalo milk were significantly higher than those in Holstein cows(p<0.01).The content of fatty acids in the three kinds of milk was the highest c16:0,which was up to 2986.31mg/100g in buffalo milk,accounting for 35.33%.In addition,the content of c18:1 reached 2419.31mg/100g,accounting for 28.62%.The beneficial unsaturated fatty acids in the two kinds of buffalo milk were also significantly higher than those in Holstein cows,in which CLA and EPA of conjugated linoleic acid were significantly higher than those in Holstein cows(p<0.01).Meanwhile,the lipid protein lactose in Italian buffalo milk and hybrid buffalo milk was significantly higher than that in Holstein cow milk(p<0.01).2.The expression levels of SCDI gene in Italian buffalo milk were significantly higher than those in Holstein cow milk(P<0.01),and the expression levels of Fas Srebp Ppary Gpam Agpat6 and Acaca related genes were significantly higher than those in Holstein cow milk(P<0.01).3.Through RNAi experiment,it was found that sirna-scd1 successfully interfered with the expression of SCD1 gene in buffalo breast epithelial cells,and the expression of SCD1 gene decreased by 41%.The results of qrt-pcr showed that after the interference of SCD1 gene,the expression levels of FAS and Ppary related to fatty acid metabolism were significantly increased(P<0.01),the relative expression levels of Srebp were increased without significant difference(P>0.05),and the relative expression levels of Acaca Agpat6 and Gpam were significantly decreased(P<0.01).After the lentivirus granule was overexpressed in SCD1,the expression level of SCD1 gene was increased by 7.8 times,the expression levels of Acaca and FAS were decreased(P<0.01),and the expression levels of Gpam Agpat6 Srebp 1 and Ppary were significantly increased(P<0.01).In the interfering group,the total amount of fatty acids decreased,the content of saturated fatty acids increased,and the content of unsaturated fatty acids decreased,among which the content of c16:0 and c18:0 increased,the content of unsaturated fatty acids decreased but was not significant,and the content of c18:2 was almost unchanged.Higher expression set the amount of fatty acids in cells,lower content of saturated fatty acids,unsaturated fatty acid content increased with C16:0 and C18:0 content reduce,unsaturated fatty acid C16:1 C18:1 and C18:2 levels rise,results and interference group,on the other hand,with the expected results agree with the experimental results show that SCD1 gene in saturated fatty acids C16:0 and C18:0,unsaturated fatty acid C16:1 C18:1 and C18:2 has important role in regulation.
