| In this paper,the Chinese herbal medicine purslane was used as the main research line.The catharsis-producing model of ESBLs-producing Escherichia coli that caused diarrhea was isolated and screened from the diarrhea intestines of piglets and other related diseases.The therapeutic mechanism of the animal model constructed by E.coli.The main methods and results of the test are as follows:1、The pathogen was isolated from the diseased piglets and identified as E.coli by 16sRNA.The virulence gene was detected by PCR,and it was found that the bacteria contained the multi-drug resistance regulatory gene marA,the anti-free radical gene sodA,the curly pilus gene csgA,the meningitis-related and temperature-regulated pili gene mat,and the lipopolysaccharide(LPS)gene R1.The virulence factors of the outer membrane protein gene ompA,type I pili(D-mannose-specific adhesion)gene fimC,serum survival gene iss,and Yersin’s iron gene fyuA.The diarrhea virulence of this E.coli may be related to R1,csgA,and fimC.2、The MIC of antibiotics and water extract of Portulaca oleracea L.was tested by double dilution method.The ICP was also tested by 1/2 MIC of water extract of Portulaca oleracea L.and MICx2 of antibacterial drug.The drug resistance gene of the strain was subjected to PCR amplification detection.The results showed that the Escherichia coli carrying drug resistance genes were:sulfonamides:sul2;tetracyclines:tetA;β-lactams:CTX-MU;plasmid-mediated quinolones:qnrD,qepA;fosfomycin-resistant:fosA3;Multi-drug resistant ESBLs-producing Escherichia coli.According to the 2018 version of the CLSI standard[This standard is the concentration of the drug resistance or the size of the inhibition zone of the four standards of drug resistance,drug resistance,sensitivity,and sensitivity after the American Association of Clinical and Laboratory Standards tests the bacteria.The criteria for the division.The standard is updated due to bacterial resistance.],the tested strains showed resistance to the above-mentioned antibacterial drugs,and the passage test showed that the water extract of Portulaca oleracea can enhance the sensitivity of the tested strain to most of the antibacterial drugs,and the MIC decreased.3、Animal model test:SD rats were randomly divided into 6 groups:Ⅰblank control group,Ⅱ model group,Ⅲ self-healing group,Ⅳ purslane low-dose treatment group,V-pursal mid-dose treatment group,Ⅵ horse High dose treatment group.The rat model of damp-heat diarrhea was constructed by high-fat diet,high-temperature and high-humidity environment and intraperitoneal injection of diarrhea-type Escherichia coli.According to the low,medium and high doses,the water extract of Portulaca oleracea L.was tested.The rats were treated with gavage.Blood routine,liver and kidney function indicators,inflammatory factors(TNF-α,IL-6,IL-17,IL-23),organ coefficient(liver,spleen,thymus)and pathological tissues of various organs in each group Learn to check.The results showed that the model group met the criteria of damp-heat diarrhea,and the model was established successfully.The systemic inflammation,blood lipid levels,immune function decreased,and histological examination showed multiple organ hemorrhage,degeneration and necrosis.Water extract of Portulaca oleracea L.through the inhibition of pro-inflammatory factors,regulation of inflammatory network function in rats,recovery and protection of immune organs,and anti-diarrhea,hypolipidemic,hypothermic effects;bleeding and degeneration of organs The situation also has some improvement;and the efficacy of purslane increases with increasing dose.It indicates that purslane has a good therapeutic effect on the damp-heat type diarrhea model.This experiment preliminarily explored the effect of Chinese herbal medicine of Portulaca oleracea on the model of rat damp heat type diarrhea constructed by ESBLs-producing Escherichia coli,and provided reference for the application of Chinese medicine of Portulaca oleracea L.to E.coli. |
