This study based on the traditional veterinary dampdeat diarrhea pathogenesis and clinical features, application of two methods to establish the rat dampheat diarrhea model with high glucose and lipid, hot and humid, enterotoxigenic escherichia coli and high glucose and lipid, hot and humid, senna. To observe the clinical symptoms, hematology, blood rheology, pathological histology, cytokines and intestinal mucosa secretion of mucosal immune protein A(SIg A) index changes of rats in each group. Comprehensive evaluation of the advantages and disadvantages of two model methods. For establishment conforms to the traditional veterinary medicine theory and clinical practice of dampheat diarrhea rat model to provide the reference. The main results were as follows:1. The two rat models clinical represented mental fatigue, addicted to lying, lazy move, hair yellow and mixed, feed and water reduction, perianal swelling and burning, loose stools, stool brown, foul-smelling, sticky around the anus and tail loose stools, tongue yellow greasy and weight loss and other symptoms, conform to the traditional veterinary dampheat diarrhea clinical characteristics description. High glucose and lipid, hot and humid and coli method was more severe diarrhea and diarrhea lasted for a long time than high glucose and lipid, hot and humid and senna method. Temperature change was due to the effect of e. coli, high glucose and lipid, hot and humid and coli method increased significantly(P<0.05) than high glucose and lipid, hot and humid and senna method.2. High glucose and lipid, hot and humid and e. coli model, compared with the blank group, the total number of white blood cells and the number of lymphocytes in the body significantly increased(P<0.01), the number of red blood cells, hemoglobin concentration, number of red blood cells deposited and platelet significantly increased(P<0.05), whole blood viscosity in the 1s-1,5s-1 and 10s-1 shear rate increased significantly(P<0.05), in the 100s-1 and 200s-1 no significant difference in shear rate, plasma viscosity in the 1s-1, 5s-1 and 30s-1 shear rate increased significantly(P<0.05), in the 200s-1 shear rate difference was not significant; High glucose and lipid, hot and humid and senna model compared with the blank group, the number of red blood cells, hemoglobin concentration, red blood cell pressure and number of platelet in the body ignificantly increased(P<0.01), the total number of white blood cells and lymphocytes significantly increased(P<0.05), whole blood viscosity in the 1s-1 and 5s-1 shear rate significantly increased(P<0.01), in 10s-1, 100s-1 and 200s-1 shear rate increased significantly(P<0.05), plasma viscosity in the 1s-1, 5s-1 and 30s-1 shear rate significantly increased(P<0.01), in the 200s-1 shear rate increased significantly(P<0.05).3. High glucose and lipid, hot and humid and e. coli model compared with the blank group, the serum IL-1β, IL-2, IL-6 and intestinal mucosa SIg A levels significantly increased(P<0.01), thymus index and spleen index increased significantly(P<0.05); High glucose and lipid, hot and humid and senna model compared with the blank group, the serum IL-1β and IL-6 levels significantly increased(P<0.01), serum IL-2 and intestinal mucosa SIg A levels, thymus index and spleen index significantly increased(P<0.05).4. Two dampheat diarrhea rat models were characterized by enlargement of the liver, gallbladder filling, rough dull surface of the liver, the color yellow, lobular necrosis, hepatic cord arrangement disorder, liver cell degeneration and necrosis. Stomach obvious blowing, mucosal edema, epithelial cell necrosis, inflammatory cell infiltration. Intestinal wall thinning, the intestinal tract is full of dark yellow lean malodorous contents, epithelial cell swelling, partial necrosis, inflammatory cell infiltration. High glucose and lipid, hot and humid and e. coli rat model gastrointestinal mucosal epithelial cells necrosis and inflammatory cell infiltration more serious than high glucose and lipid, hot and humid and senna model.In conclusion, high glucose and lipid, hot and humid and enterotoxigenic escherichia coli and high glucose and lipid, hot and humid and senna methods can be successfully established the dampheat diarrhea rat model. Rat clinical manifestation such as mental fatigue, perianal swelling and burning, loose stools, fecal stench and slimy yellow tongue fur etc., and the significant increase of whole blood viscosity, plasma viscosity, spleen index and thymus index, IL-1β, IL-2, IL-6, SIgA and the gastrointestinal hepatic obvious pathological changes. High glucose and lipid, hot and humid and e. coli method mainly cause severe rat inflammatory response and gastrointestinal mucosal injury. High glucose and lipid, hot and humid and senna method mainly cause severe diarrhea and blood viscosity of rat.
