Lily is a bulbous flower with high economic value,derived from Lilium.It ranks fourth among the top ten cut flowers and ranks among the top three in China’s flower industry.Although China has abundant wild lily resources,the new varieties cultivated each year are far from the Netherlands.It is an important content to promote the development of lily industry by using the existing lily varieties for breeding,or hybrid breeding with our abundant wild lily resources to cultivate new lily varieties with independent intellectual property rights.The odd-allotetraploid lily “Honesty” which contains a set of Longiflorum genomes and three sets of Asiatic genomes.In current lily breeding research,this type is special.It is of great significance for lily breeding to explore the fertility of lilium odd-allotetraploid.In this study,the odd-allotetraploid lily “Honesty” was selected as the female parent.After observing its meiotic behavior,it was hybridized with three diploid Asiatic lilies ?Black Out?,?Lollypop?,?Tiny Padhye? five Tetraploid Asiatic lilies ?090024-16?、?Pearl Stacey?、?Pearl Melanie?、?Pearl Jennifer?、?Pearl Carolina?,and their progenies were analyzed by cytology.The results were as follows:1.The meiosis observation of the “Honesty” revealed that there were partially lagged chromosomes at telophase II and tetrad and there were abnormal phenomena such as unequal division.Pollen germination rate was only about 6.3%..2.The results of pollen germination of 8 Asian lily parents showed that the germination rate of three diploid Asian lily was between 37.3% and 42.3%,and the five tetraploid Asian lily was between 33.6% and 40.2%.The germination rate of Asian lily materials were better and it could be used as male parent to participate in the hybridization.3.In this study,three LAAA×AA hybrid combinations were selected,We polled 60 flowers and harvested 6 fruits,and then obtained 15 seedlings through embryo rescue;we selected five LAAA×AAAA hybrid combinations.50 flowers were polled.we harvested 46 fruits,and then obtained 594 seedlings through embryo rescue.It indicated that the odd-allotetraploid ’Honesty’ could be used as a female parent and had a good compatible with tetraploid Asiatic lilies.4.We used GISH technology to analyze 17 hybrid progenies..The chromosomes number in 8 progenies of LAAA×AA varied from 34 to 72,their average was 49.63.The number of L was between 1 and 10 and L/A(recombinant chromosomes)was between 2 and 4.While the chromosomes number in 9 progenies of LAAA × AAAA varied from 43 to 65,their average was 49.59.The number of L was between 2 and 7,and L/A was between 2 and 4.All progenieswere aneuploidy.They had variable L and L/A.The average number of chromosomes in progenies was close to tetraploid(2n=4x=48).To sum up,the odd-allotetraploid lily could be used as a female parent to hybridize with a suitable diploid or tetraploid Asiatic lily.‘Honesty’ meiosis produced aneuploid gametes and formed a new genome reorganization.which had great significance for the introgression breeding.
