Chitosan acyl thiosemicarbazone derivatives were prepared by localization protection of Chitosan and thiosemicarbazone.Through the combination of the two compounds,it not only retains the better antiviral and antibacterial activity of thiosemicarbazone,but also reduces the toxicity and improves the antioxidant activity.Sixteen kinds of Chitosan acyl thiosemicarbazone derivatives were prepared and screened by our research group.The results showed that they had strong antioxidant activity and antibacterial activity in vitro.We hope to prepare low toxicity and high efficiency pesticides and veterinary drugs by this method.The effects of five concentrations of Acyl-thiosemicarbazone-Chitosan derivatives(0.005?0.015?0.025?0.035 and 0.045 g·mL-1)on the lifespan,and fecundity of Drosophila melanogaster have been investigated.Sixteen Acyl-thiosemicarbazone-Chitosan derivatives?five thiourea crystals?high molecular Chitosan and low molecular Chitosan were measured.The results showed that compared with the thiourea crystal,the toxicity of most derivatives is greatly reduced.The seven derivatives(3,6-DA(p-T)TSCZHCS?DA(o-T)TSCZHCS?DAPTSCZHCS?3,6-DCA(o-T)TSCZHCS?3,6-DCA(p-NP)TSCZHCS?3,6-DBZ(o-T)TSCZHCS?3,6-DBZPTSCZHCS)had the least effect on the longevity and fecundity of Drosophila melanogaster,showing the characteristics of low toxicity,strong fecundity and good antioxidant activity.All derivatives had a certain degree of influence on the lifespan and reproductive capacity of Drosophila melanogaster in the concentration range of 0.005-0.045g·mL-1.For male Drosophila melanogaster,according to the life-span test of Drosophila melanogaster.When the concentration of Dibenzoylated phenyl-thiosemicarbazone-Chitosan derivative(3,6-DBZPTSCZHCS)is 0.035 g·mL-1,the average life,half-life and longest life all reach the peak 67.2±4.0?47.9±5.9 and 85.4±5.0 days.For female Drosophila,according to the life-span test of Drosophila melanogaster,the Dibenzoylated phenyl-thiosemicarbazone-Chitosan derivative(3,6-DBZPTSCZHCS)had little difference in the concentration range of the experiment(0.005 g·mL-1-0.045 g mL-1).The peak values of average life span,half life span and longest life span were 70.3±5.5?58.7±6.1 and 84.9±7.0 days respectively,which were significantly better than that of the control group PTSCZ.In the range of 0.015 g·mL-1-0.045 g mL-1,it was also significantly superior to Chitosan(HCS)and prolonged the longest life span of female Drosophila melanogaster.According to the fecundity experiment of Drosophila melanogaster and considering the F1 and F2 of the fecundity experiment of Drosophila melanogaster,the inhibitory effect of three Diacetylated phenyl-thiosemicarbazone-Chitosan derivativees(3,6-DA(p-NP)TSCZHCS?DA(p-T)TSCZHCS)on the reproduction of Drosophila melanogaster can reach the optimal value.For F1 generation,the toxicity of DA(p-T)TSCZHCS was better than that of control group(p-T)TSCZ crystal in the range of concentration tested.At the concentration of 0.045 g mL-1,the inhibition rate of the fecundity of Drosophila melanogaster reached a minimum of 10.31±1.71%.At the same time,the inhibition rate of DA(p-T)TSCZHCS was almost the same as that of Chitosan raw material(12.64±1.51%)at the concentration of 0.025 g.mL"1.For F2 generation,the inhibition rate of 3,6-DCA(p-NP)TSCZLCS on the fecundity of Drosophila melanogaster reached the minimum of 11.34±1.19%at 0.015 g mL-1 concentration,and the inhibition rate of LCS was 8.66±0.56%at the same concentration.The toxicity of 3,6-DCA(p-NP)TSCZLCS increased when the concentration was lower or higher.This study provides an experimental basis for the further development of Chitosan derivatives with high antioxidant activity and low toxicity,and lays a foundation for the preparation of new environmental friendly pesticides and veterinary drugs.
