Royal jelly is a bee-secreting product with rich nutrition and efficacy.According to some studies,royal jelly can regulate cell metabolism by increasing insulin pathway,rapamycin complex 1(TORC1)activity,and promote larval development into royal jelly.Drosophila melanogaster is a good model to study the active components in royal jelly,and many kinds of royal jelly active components can play a role in Drosophila melanogaster.The experiment was carried out by using honeybees as experiment and fruit flies.TORC1 is a key factor regulating cell metabolism and cell growth,and its increased activity is closely related to diseases such as aging and tumor.Nprl3,as a GATOR1 component protein,can inhibit TORC1 activity.we used the previously established nprl3 mutant flies as a model to detect the salvage effect of royal jelly on the phenotypes of nprl3 mutant flies caused by dysregulated TOR activity,poor antioxidant capacity and low lipid content using western blot,antioxidant level detection,immunofluorescence and transcriptome sequencing to elucidate the regulatory effect of royal jelly on fly metabolism.And identify the main active components in royal jelly which can save nprl3 mutation flies.provide theoretical guidance for the health care use of royal jelly in TOR pathway disorders related diseases.this study found that feeding royal jelly can significantly increase the survival rate of nprl3 mutant flies.through heat treatment,protease digestion,we found that the protein components in royal jelly play an important role in improving the survival of nprl3 mutant flies.Royal jelly can increase the activity of antioxidant enzyme SOD and CAT in nprl3 mutant flies,reduce the content of peroxide in vivo;feeding royal jelly can also increase the content of triglyceride(TG)and adenosine triphosphate(ATP)in vivo,and improve the defect of fat storage and utilization in nprl3 mutan flies.the detection of phosphorylation levels of TORC1 downstream proteins revealed that royal jelly treatment could reduce TORC1 activity in nprl3 mutation fies.these findings suggest that royal jelly improves metabolic defects caused by elevated TORC1 activity,nprl3 mutant flies is a good model for studying active bioactive components in royal jelly.
