In this study,the CPV puppies model was constructed by the method of fecal toxicity.The experimental dogs were divided into the standard treatment diet group(STD)and the standard treatment+fecal microbiota transplantation group(STD+FMT).The stools and blood samples of the two groups of dogs at different time points before and after the onset were observed.The therapeutic effect of FMT on CPV was observed and evaluated from the aspects of viral load,blood index and intestinal microecology.The main research work and results are summarized as follows:1)Therapeutic effect of fecal microbiota transplantation on canine parvovirus disease.The changes of body mass index and disease activity index of the STD and STD+FMT groups of dogs at different time points before and after the onset were recorded.The body mass index of the two groups of dogs decreased after the onset all in 4d.At the lowest time,the body mass index of the puppies in the STD+FMT group recovered faster than that in the STD gro up.The DAI scores of the two groups showed significant differences on the 3?4?5 and 6 after onset,and the disease activity scores of the dogs in the STD+FMT group.It was always lower than the STD group;the blood index and C-reactive protein were detected in the puppies.The results showed that the white blood cells(WBC)of the two groups of dogs were continuously decreased three days before the onset of the disease,rising from the fifth day,and the third after the onset.In the 5th day,the WBC of the STD-FMT group was significantly higher than that of the STD group;the C-reactive protein reached the highest value on the first day of the disease but there was no significant difference,and then began to decline.On the 3rd and 5th day of the onset,the STD+FMT group The CRP of the experimental dogs was significantly lower than that of the STD group.The virulence of puppies was detected by the established CPV fluorescence quantitative PCR method.The results showed that the viral load of the dogs in the STD+FMT group decreased rapidly.Intestinal histopathological sections showed that FMT treatment can alleviate intestinal tissue damage caused by CPV.2)Effects of fecal microbiota transplantation on intestinal microecology in canine parvovirus patients.The article collected the fecal samples from different groups of experimental dogs at different time points before and after the onset,extracted the total DNA for 16S rRNA PCR amplification,and used high-through put technology to analyze the effect of fecal transplantation on the intestinal micro-ecology of dogs.The results showed that the diversity and abundance of intestinal flora decreased significantly after CPV infection.FMT could significantly increase the diversity and richness of intestinal flora in dogs.The results of PCoA and NMDS analysis showed that after 7 days of FMT treatment,dogs with STD group were tested.In contrast,the composition of intestinal flora in the STD+FMT group was clo ser to that before the experiment(HD);the statistical results of species classification showed that after 7 days of treatment,The statistical results of species classification showed that after 7 days of treatment,compared with the STD group,the relative abundance of Firmicutes and Bacteroidetes in the intestinal micro flora of the S TCD+FMT group was significantly increased,and the relative abundance of Proteobacteria was significantly decreased.At the genus level,after FMT treatment,the Escherichia of the STD+FMT group was significantly lower than that of the STD group,and there was no significant difference compared with the HD;the relative abundance of Lactobacillus,Faecalibacterium and Bacteroides was significantly increased,and the relatively abundance of Blautia was decreased slightly.After 7 days of FMT treatment,the intestinal flora relative abundance at the phylum and genus level of the STD+FMT group was closer to that before the experiment.In summary:the use of fecal microbiota transplantation for CPV puppies can alleviate the disease and accelerate the recovery of body mass,eliminate systemic inflammatory response,promote the body to clear the virus,while improving intestinal micro-ecology and restoring the intestinal barrier.
