Plant U-box protein(PUB)is an important component of ubiquitination system responsible for recognizing substrate proteins,and is involved in plant resistance reaction,signal transduction and transcriptional regulation.In order to clarify the anti-nematode-related PUB protein genes(Gm PUBs)in the interaction between soybean and soybean cyst nematode(SCN).In this paper,the susceptible variety Liaodou 15 and the resistant variety gray-skin black bean were inoculated with soybean cyst nematode No.3(SCN3),and Changes in expression levels of Gm PUB1,Gm PUB5,Gm PUB6,Gm PUB7,Gm PUB8 and Gm PUB9 in Gm PUBs family after infection with susceptible strains by cystic nematode by q RT-PCR.Study whether these six genes are involved in the disease resistance of soybean to cyst nematode.In order to clarify the effects of SA and JA signaling substances on the expression of Gm PUBs,the expression levels of these six genes were measured by exogenous hormones SA and Me JA,respectively.The number of these genes was measured at different ages.Whether to participate in SA or JA anti-nematode signaling pathways.The main results are as follows:1.The results of Gm PUBs gene expression test in soybean indicate that Gm PUB1 and Gm PUB6 are related to resistance in soybean and cyst nematode interaction,and Gm PUB8,Gm PUB5 and Gm PUB9 may be involved in the regulation of soybean anti-cytoknot development.The resistant varieties were inoculated into the 3rd,3d,5d,10 d and 15 d races of soybean cyst nematode 1d,3d,5d,10 d and 15 d.The expression of 6 Gm PUBs genes was determined by q RT-PCR.In the resistant varieties,Gm PUB1 and Gm PUB6 were inoculated with SCN3.Afterwards,it is always significantly up.Among the resistant varieties,Gm PUB8 was significantly up-regulated after inoculation with SCN3 for 10 d and 15 d,and Gm PUB5 and Gm PUB9 were significantly up-regulated after inoculation with SCN3 for 15 d.2.According to the change of GmPUBs gene expression after soybean treatment,it was confirmed that Gm PUB6 and Gm PUB8 are involved in SA anti-SCN developmental signaling pathway.Continuous application of SA in three days after resistance to soybean leaves inoculated with SCN3,Root numbering showed no significant difference in J2 invasion.The number of J3 and J4 nematodes in roots decreased significantly after inoculation with SCN3 for 10 d and 15 d,indicating that SA can inhibit the development of SCN in roots..The expression of gene expression was detected by q RT-PCR.It was found that in the resistant varieties,the gene expression of Gm PUB6 and Gm PUB8 after SA treatment and inoculation with SCN3 for 10 d and 15 d was significantly up-regulated compared with the treatment with SA and SCN3 only.3.According to the change of Gm PUBs gene expression in soybean treated with Me JA,Gm PUB1,Gm PUB7 and Gm PUB9 were involved in JA anti-SCN invasion signaling pathway.Continuous application of Me JA to SCN3 was induced in three days after resistance to soybean leaves.Root staining showed that the number of J2 in roots decreased significantly after inoculation of SCN3 for 1d and 3d,indicating that JA inhibited the invasion of SCN.The expression of gene expression was detected by q RT-PCR.It was found that in the resistant varieties,Gm PUB1,Gm PUB7 and Gm PUB9 were significantly up-regulated after Me JA treatment and SCN3 inoculation for 1d and 3d,and the ratio of Me JA treatment and SCN3 only.
