The duck industry is a traditional breeding industry in China.The rich duck products are very important for people's pursuit of high quality life.However,in the production,the duck industry often faces the risk of disease outbreaks.In the process of prevention and control of bacterial diseases,it is a highly desirable way to improve the clinical deficiencies by researching and developing new dosage forms of drugs.Thiamphenicol is a broad-spectrum antibacterial agent widely used in the veterinary clinic for the prevention and treatment of digestive tract and respiratory diseases in livestock and poultry.With the development of pharmaceutics,micelles have received extensive attention as an effective drug carrier.The application of polymer micelle technology to thiamphenicol can overcome the deficiency of thiamphenicol in clinical treatment.In this study,the sulforamycin micelles were prepared,and their physicochemical properties were analyzed.The metabolic kinetics of Thiamphenicol micelles in ducklings was further explored.The clinical research and development of a new medicinal dosage form of Thiamphenicol.Lay the foundation of the previous period.The main research contents are as follows:The detection of preparation of thiamphenicol micelles is carried out by thin film dispersion method and high performance liquid chromatography.This method has good selectivity,high separation efficiency,high detection sensitivity and wide application range.Taking the encapsulation efficiency and drug loading as the indicators,the thiophenemycin micelles were investigated by single factor,Determine the main factors affecting the amount of organic solution,the amount of thiamphenicol,the volume of hydration medium and the pH of the buffe and thiamphenicol micelles was optimized by the Box-Behnken Design response surface to obtain the optimal prescription: 40 mg thiamphenicol,The volume of methanol solvent was 20 m L,phosphate buffer solution(PBS)pH 8.0,and PBS hydration volume 15 m L.The physicochemical properties of thiamphenicol micelles and the metabolic kinetics of ducks were investigated.The physicochemical properties of thiamphenicol micelles were investigated from four aspects.The grain size of thiamphenicol micelles was 112 nm,with a regular structure,similar to spherical shape,uniform micelle morphology,and a Zeta potential of-23.4 mv.Thiamphenicol micelles have high stability at 4°Cand 8 months and poor stability at room temperature.The pharmacokinetic characteristics of thiamphenicol micelles in healthy ducks were studied by intravenous administration.The healthy ducks were injected with a single dose of intravenous injection of thiamphenicol micelles at a dose of 10 mg/kg b.w.,and blood was collected from heart 20 times in 96 hours.The blood concentration was determined by high performance liquid chromatography.The results showed that the intravenous injection of thiamphenicol micelles conformed to the non-absorbed two-compartment open model,the central chamber distribution volume(V(c))was 0.618 L/kg,and the distribution half-life(T1/2?)was 0.496 h,eliminating half-life(t1/2?)was 5.023 h,and the body clearance(CL(s))was 0.313 L/kg/h.The results showed that after intravenous injection of thiamphenicol micelles,the thiamphenicol micelles were rapidly absorbed,widespread,eliminating half-life extension in the duck vivo.The relative bioavailability of thiamphenicol micelles is relative high.In summary,micelles can significantly improve the solubility and bioavailability of thiamphenicol,and the clinical deficiencies of thiamphenicol can be changed by new medicinal dosage form.Micellar is expected to be an ideal carrier for thiamphenicol.
