Pudong white pig is a local pig breeds that from Pudong and Fengxian districts of Shanghai.It is bred long-term by local farmers and advantages of high reproductive rate,delicious meat.As the only white pig breeding of China,Pudong white pig is great value in the protection of genetic resources.But Pudong white pig was on the verge of disappearing because it is not suitable for industrialized farming.Now the industrialized aquaculture exposes more and more problems such as drug abuse,infectious disease is prevalent and the decrease of meat taste.It will be an effective approach to deal with these problems by using the pig breeding resources from various regions of the country and mainstream breeds in large farm to breed different new pig breeds.Carrying out research Of Pudong white pig on genetic characteristics and resistance to disease can help us to use its genetic resources better,can also help protect the provenances of Pudong white pig.Mx1 gene is associated with the resistance of animal to the influenza virus and the vesicular stomatitis virus,and Nramp1 gene is associated with the resistance of animal to the Intracellular pathogen infection disease.These 2 genes became the more popular candidates for disease resistance genes in recent years.Many studies at home and abroad have proved that the mutations of Mx1 gene exon sixth and Nramp1 gene fourteenth intron have effect on disease resistance of pigs.Studying on two gene fragments and related natural resistance of Pudong white pig this will help to understand the genetic characteristics and related performances of Pudong white pig and can also promote the antiviral effects research of Mx1 gene and NRAMP1 gene.Random sampling of Pudong white pig populations to amplify and gene sequence the fragments of Mx1 gene exon 14 and Nramp1 gene intron 6.The results shows that there are many mutations in the 2 gene fragments of Pudong white pig,most of the mutations are found not in the other pigs:1,The mutations found in the exon 14 of the Mx1 gene sequence :C47?missing,A49?missing,C54?missing,A63?missing and A76?G and T146?C,total 6 mutation sites.2,The mutations found in the intron 6 of the Nramp1 gene sequence :A269?G,G363?A and 1or2 thymines inserted after 29 bp,total 3 mutation sites.Random sampling and detecting the selected genes related immunological indexes of Pudong white pig and bigwhite pig populations on the same farm.ELISA testing reports shows that the levels of serum interleukin 4,Interleukin 12 and Interferon-? in two pig breeding populations were not significantly different(P > 0.05).Pudong white pig is a very characteristic local pig breeds.There are lots of distinctive mutations in the genome relative to other pig breeds.But there is no special of Pudong white pig in natural disease resistance on the immunological indexes in the detection results.Preliminary research results indicate that Pudong white pig is significantly different with big white pig on mutation sites of Mx1 gene extron 14 and NRAMP1 gene intron 6,but there is no significant difference in the related immune performance of Pudong white pig and big white pig,and similar studies in Chinese local pig breeds is rarely reported.These findings can provide some basic data for the future of Pudong white pig natural disease resistance research and research in gene breeding,also for clearer understanding of MX1 gene and NRAMP1 gene resistance to disease.
