The whitebacked planthopper is one of the main pests of rice and its damage is very serious.It involves a large area and a wide area.White-backed planthoppers are migratory pests and many of the factors affecting their wing-type differentiation are complex,including juvenile hormones.Dinotefuran is a new generation of nicotinic insecticides as an important agent for the control of rice planthoppers.However,at present,reports of studies on juvenile hormones such as whitebacked planthoppers,dinotefuran and their effects on whitebacked planthoppers are relatively rare.Therefore,this paper studies the effects of dinotefuran on the juvenile hormone and related genes in the treatment of the planthopper.There are several ways.1.using enzyme-linked immunoassaymethod(ELISA)to detect the white-backed planthopper insect different age stages juvenile hormone ? levels in the body.2.The gene length of the genes involved in the synthesis of juvenile hormone related genes by the race technology clone and the white back planthopper,and the full length of the genes involved in the decomposition of juvenile hormone.3.The expression levels of SfJHAMT and SfJHEH in different life stages were detected by real-time fluorescence quantification.4.Treatment of white-back planthoppers with the lethal dose of dinotefuran,to detect the expression of SfJHAMT and SfJHEH at different stages of different sublethal dose under different sublethal dose under different sublethal doses was detected by real-time fluorescence quantification.After the experiment,the following results are obtained.1.White-backed planthopper nymphs during 5 age juvenile hormone ? content is the lowest,Adult stage white-backed planthopper juvenile hormone ? increases with age.The content of female adult of 5 days reached 201.4 ng/L.There was a decline after 7 days.Similarly,there was no significant difference in the content of juvenile hormones between different days,but the content of juvenile hormone was higher than that in the age of 5 years.2.The juvenile hormone methyltransferase SfJHAMT is expected to have a full length of 1247 bp,an ORF region of 230-1057 bp and a length of 828 bp,which is expected to encode 275 amino acids;the juvenile hormone epoxide hydrolase SfJHEH is expected to have a full length of 2139 bp,an ORF region of 2616-1616 bp,a length of 1356 bp,and is expected to encode 451 amino acids.3.By fluorescence quantitative detection of the expression of two genes in different stages of wormwood.Found SfJHAMT and SfJHEH not only in white-backed planthopper nymphs and adults of expression is different in different life stages,and expression in male and female gender and the gender is male and female mating is different under different conditions.4.The expression of SfJHAMT and SfJHEH at different stages of wormwood was tested by using futiamine-sublethal dose to deal with white back planthopper.It was found that SfJHAMT and SfJHEH gene expression were different in the same life stage at different concentrations.And the expression levels of SfJHAMT and SfJHEH were also different in different life stages.
