The white backed planthopper[Sogatella furcifera(Horváth)]is a major migratory pest of rice crops in Asia.The flight muscles of the insect are the "engine" that provides a powerful driving force for their flight.It is of great significance for their migration.At present,the chemical is the most effective means for the prevention of migratory pests.Dinotefuran is a third-generation neonicotinoid has the feature of high efficiency,low toxicity,a wide range of control and high safety and so on would give it a broad future of application.In recent years,the effect of insecticides on pests is a hot research direction.Some studies have shown that the ultrastructure of flight muscle of some insects will be affected by some chemical agents.However,the effects of insecticides on the flight muscles of the planthopper have not been reported.Based on this background,in this paper,measuring the female sogatella furcifera(Horvath)flight muscle ultrastructure and the fiber diameter,cross-sectional area of myofibrils,sarcomere length,mitochondria than these indicators to explore the effect of total dinotefuran on its flight muscle ultrastructure.To investigate the effect of dinotefuran on the apoptosis of flight muscle by measuring the total muscle DNA and total protein content.Use the TUNEL detect the degradation of its flight muscles to investigate the effects of the two insect larvae on the degradation of its flight muscles.The main results are as follows:(1)On the 1?3 day after emergence of S.furcifera,the cross-sectional area of the myofibrils of untreated group ranged from 1.15 to 2.01 ?m2,the diameter of myofibrils ranged from 1.15 to 2.03 ?m,the sarcomere length ranged from 1.91 to 2.66 ?m,them were increased with eclosion days.The percentage of mitochondria in flight muscle fibers ranged from 31.87%to 38.53%,and it initially increased and then decreased with the increase in the eclosion days.The cross-sectional area of the myofibrils and the diameter of myofibrils of dinotefuran-treated group increased with the eclosion days and greater than that of the untreated group.On the first day after emergence,the higher the concentration of dinotefuran,the greater the increase.On the second and third day after emergence,the cross-sectional area of the myofibrils and the diameter of myofibrils of the LC20 dinotefuran-treated group is the most we measuring result.The sarcomere length of the dinotefuran-treated group initially decreased and then increased with the increase in the eclosion days.On the first day,the higher the concentration of dinotefuran,the less the decrease.On the third day,the higher the concentration of dinotefuran,the greater the increase.The percentage of mitochondria in flight muscle fibers of the LC10 and LC20 dinotefuran-treated group increased with the eclosion days.The LC10 dinotefuran-treated group is the most we measuring result.The LC30 dinotefuran-treated group was changing very rapidly,it initially decreased and then increased compare with untreated group.The results showed that the ultrastructure of the flight muscle of the female white planthopper was enhanced under the concentration of the stress of the dinotefuran.(2)On the 2-5 day after emergence of S.furcifera.The content of the total DNA,total protein in the flight muscles of females S.furcifera initially increased and then decreased with the increase in the eclosion days.On the third day of S.furcifera emergence,the total DNA,total protein in the flight muscles is the most we measuring result on the untreated group.On the fourth day of S.furcifera emergence,the content of the total DNA,total protein in the flight muscles is the most we measuring result on the dinotefuran-treated group.The content of the total DNA,total protein initially increased and then decreased with the increase of the concentration of dinotefuran.The content of the total DNA,total protein of the LC20 dinotefuran-treated group is the most we measuring result was significantly greater than the other group.The results showed that the content of the total DNA,total protein was enhanced under the concentration of the dinotefuran.The time of apoptosis was postponed.(3)TUNEL detection showed that on the fourth day of the S.furcifera emergence,the apoptotic cells were detected in the flight muscle with untreated group.on the fifth day of the S.furcifera emergence,the apoptotic cells were detected in the flight muscle with dinotefuran-treated group.The results showed that the time of the development of flight muscle was extended.The time of apoptosis was postponed.The time of turn to procreation was postponed.So,the time of the migration of the S.furcifera was extended.
