| Dermatophytosis is one of the main diseases of pet all over the world,and the major cause is associated with fungi infection,such as genera Microsporum,Trichophyton and Epidermophyton.This diseases characterized mainly invading infections,repetition,therapeutic period.Currently,there are many chemically synthesized anti-dermatophytic drugs which are often associated with resistance and significant side effects.It is important to develop anti-fungal Chinese medicine or natural medicine.In this study,We have success studyed Galla chinensis solution(GCS),we make a research for the activity of GCS against dermatophytosis in dogs.The main results are as follows:1 Establish dogs model of dermatophytosisDogs were randomized into three groups,include Microsporum infection group,Trichophyton infection group and Epidermophyton infection group.The skin of these dogs on both sides of the back four parts was clipped by an electric shaver(2.5 cm 2.5 cm)under anesthesia,respectively.0.1 mL of four different concentrations conidia solution was smeared on this area.The results showed that the concentrations of the fungi were determined as 1 10~6 CFU/mL,5 10~5 CFU/mL and 1 10~6 CFU/mL for M.canis,M.gypseum and T.mentagrophytes,respectively.The skin of inoculated can be seen erythema,herpes,scaly and white scab,and mycology tested was positive.2 Efficacy of GCS treatment of dermatophytosis in experimental clinical trials50 dogs have confirmed dermatophytes were randomized into five groups(ten animals for each group):(i)Untreated group,(ii)high doses of GCS group,(iii)medium doses of GCS group,(iv)Low doses of GCS group,(v)Positive control(Compound Tujingpi Tincture).10 dogs as control group.GCS and Compound Tujingpi Tincture were applied topically to the infected area once a day for 14 days.In T.mentagrophytes model,M.gypseum model,M.canis model,the mycological cure rates of high doses of GCS were60%,80%,80%,respectively.The mycological cure rates of medium doses of GCS were40%,60%,60%,respectively.The mycological cure rates of medium doses of GCS were20%,20%,20%,respectively.The mycological cure rates of Compound Tujingpi Tincture were 30%,20%,20%,respectively.The total efficiency of high and medium of GCS were100%.The total efficiency of low-dose groups were 90%,90%,80%,respectively.The total efficiency of Compound Tujingpi Tincture were 90%,80%,80%,respectively.The results showed that GCS is a beneficial antifungal agent in vivo for dermatophytosis and GCS has no effect on the blood of the dog.3 Efficacy of GCS treatment of dermatophytosis in expanded clinical trials120 pets had confirmed dermatophytes which were randomized into two groups,includeing the positive control group and the Galla chinensis solution group.The drugs were applied topically to the infected area once a day for 14 days.The mycological cure rates and total efficiency of GCS were 60%and 86.7%,respectively.The mycological cure rates and total efficiency of Compound Tujingpi Tincture were 50%and 73.7%,respectively.The results showed that GCS was effective for dermatophytosis,and it could be used as an efficient drug for treatment of topical fungal infections.Above all,it is feasible to establish a model of skin fungal infection in dogs and the5%GCS is best for clinic use.The mycological efficacy rates were 40%,40%,60%,respectively and total efficiency of rates of were 100%,which is higher than Tujingpi Tincture on days 14 post-treatment.On this basis,120 dogs were used for expanded clinical trials.The results showed that the cure rates and total efficiency of GCS(60%and86.7%)were greater compared to the positive group.In conclusion,Galla chinensis solution is a potent antifungal agent in vivo for dermatophytosis.It could be used for treatment of topically fungal infections. |
