Cryptosporidium spp.and Enterocytozoon bieneusi are two important species of parasitic diseases and are the main pathogens causing cryptosporidiosis and microsporidia in livestock and humans.The two species of parasite can cause self-limiting diarrhea for the host of normal immunity,nonetheless,all can be cured under treatment,but for immunocompromised hosts,especially those with immunodeficiency disease,they will cause severe fatal dehydration and diarrhea.In this experiment,the parasitic spores were collected by floating sucrose solution,and the oocyst DNA was extracted and then subjected to nested PCR amplification and sequencing.The fecal samples from the yak farm in Aba area of Sichuan were tested for Cryptosporidium and Enterocytozoon bieneusi,and then align sequence analysis.In this study,the method of nested PCR amplification of ITS gene was combined with the sequencing results,and the genotypes of Enterocytozoon bieneusi were detected in fecal samples from four yak farms in Heishui County and Songpan County of Aba Prefecture.And identification,the infection rate is 5%.Enterocytozoon bieneusi was detected only in diarrhea fecal samples.Two genotypes of Enterocytozoon bieneusi,BEB3 and PtEb XI,were detected in this experiment,all of which were Group 2 Genotype,this group of genotypes has host specificity,is only infectious to a single species of host,and has no zoonotic potential.In this study,nested PCR amplification of 18 S rRNA gene was combined with sequencing results to analyze and identify Cryptosporidium species from four yak farm manure samples from Heishui County and Songpan County of Aba Prefecture..Cryptosporidium infection was detected only in the feces of yak,and it was identified as a bovine sporozoite species susceptible to bovine infection(100% homology with the GenBank database sequence KF128742),with an infection rate of 0.5%;It indicates that yak cryptosporidiosis in some areas of Aba Prefecture is caused by Cryptosporidium bovis,and yak is more susceptible to it.By applying this method and integrating the analysis of Cryptosporidium and Enterocytozoon bieneusi genotypes,there is a preliminary understanding of the infection of two parasites in some areas of Aba,which will be a better study of scholars in the future.The data base of Cryptosporidium and Enterocytozoon bieneusi epidemiology was laid,and the theoretical basis for the prevention and control of Cryptosporidium bismuth and Enterocytozoon bieneusi in Aba area was provided.
