| Brucellosis is a zoonotic disease that causes infections in the body.The epidemic has caused serious economic losses to the animal husbandry and poses a great threat to the personal safety of the farmers and their related industries.Amber plate agglutination test initial screening,tube agglutination test confirmed is the most widely used Brucellosis detection method in China’s grassroots testing institutions,turbidimetry as a test method for tube agglutination test results,relying on the visual observation of the test personnel,the test results will Subject to experience levels and subjective bias.(1)In order to judge the results of tube agglutination test more objectively and accurately,this study firstly scanned the turbid tube suspension of the tube agglutination test using four spectra of 405 nm,450 nm,490 nm and 630 nm.The OD value obtained from the spectral scan determines the optimal spectrum as a unified scan spectrum for subsequent studies.Furthermore,according to the"Blood Brucellosis Diagnostic Technique"(GB/T18646-2002)tube agglutination test operation method,72 known agglutination titers were detected,and the difference between the spectrophotometric method and the turbidimetric method was compared.The results show that the OD value of the turbidity tube is the largest in the 405nm spectrum,and the OD value is significantly different from other spectral scans(P<0.05).The linear relationship determines the coefficient R~2=0.9992,and the reliability is the highest.The 405nm spectrum is determined as the unified scanning spectrum of the subsequent study.The accordance rates between 72 samples of serum titers and known titers were 83.33%(60/72)and 88.89%(64/72)(P>0.05),by using spectrophotometry and turbidimetry respectively.The concordance rate was 94.44%,and the Kappa value was0.77>0.75,indicating that the results of the two methods are consistent.The titer of the 72samples,54 positive samples,and 18 suspicious samples determined by spectrophotometry was 5.6%,3.7%and 11.1%higher than the turbidimetric method,and the spectrophotometric method was more suspicious than the turbidimetric method.The positive number of samples was 33.33%,indicating that the results of the tube agglutination test by spectrophotometry can improve the titer of the sample to be tested.(2)From 2017 to 2018,this method was used in the detection of brucellosis in Hantai District.The SAT results of positive samples detected by RBT screening were determined.The samples retained the original detection data on qualitative and quantitative tests.The results are objective and unique.The results showed that the results of five blind samples in the comparison of the testing capabilities of the veterinary laboratory in Shaanxi Province in Shaanxi were completely consistent with the reference results given by the Shaanxi Provincial Animal Disease Prevention and Control Center in 2017.The field positive samples were detected in 2018,and the turbidimetric method was obvious.Qualitative samples were determined by spectrophotometry and consistent with the results of turbidimetry.For the samples with multiple probabilities determined by turbidimetry,the results were determined by spectrophotometry.This examination provides strong data support for the spectrophotometric method determined results of tube agglutination test that from it was established to the application in the detection of brucellosis in the promotion and application of brucellosis surveillance. |
