Armillaria mellea is a facultative parasitic fungus of Basidiomycotina,Agaricales,Tricholomataceea,Armillaria,which widely distributes in forest areas.The fungi have a high medicinal value.Gastrodia elata,a precious traditional Chinese medicinal plant,is symbiosis with Armillaria,which can only grow and reproduce by providing nutrition.When Gastrodia elata is cultivated for many generations,Armillaria will meet with a significant degradation and variation.Once we choose the degraded strains symbiosis with Gastrodia elata,it can lead to sharply reducing its yield and quality.Thus in this experiment,wild Armillaria mellea was collected from the main producing areas of Gastrodia elata and excellent Armillaria mellea was introduced from the company.The collected strains were observed and counted in the laboratory and co-planted with Gastrodia elata in the field.The dominant strains suitable for Gastrodia elata planting were screened out and the genetic diversity of the dominant Armillaria mellea strains was studied.Finally,it provides a theoretical basis for the development of Armillaria mellea resources and the screening of dominant Armillaria mellea strains suitable for planting Gastrodia elata.The strains GZZJ,GZDF-1 and AHJZ with excellent biological characteristics grew fast,the fungal cords were strong,and the fungal cords had many branches,and the effect of companion cultivation was the best.The growth rate of Armillaria mellea strains was positively correlated with its effect on Gastrodia elata yield.The main results are: 1)amongour 16 strains,GZDF-1,AHJZ,GZZJ,GZHZ and JLTM showed high activities.,2)the comparative analysis of Main Agronomic Traits of Gastrodia elata showed that ring spacing,number of dotted transverse rings,arrow bud length,tuber length,tuber width,tuber thickness,umbilical cord diameter,tuber length-width ratio had little difference,and it belonged to relatively stable traits.From the total content of gastrodin and p-hydroxybenzyl alcohol in Gastrodia elata,the quality of Gastrodia elata planted with A.mellea strain AHJZ was the best,followed by A.mellea GZDF-1,GZZJ,JLTM and GZHZ.DNA sequence analysis of Armillaria showed that: The DNA sequence of strain AHJZ had a relative high similarity with that of A.sinapina strain(83.03%).The DNA sequence of strain GZZJ had the highest similarity with that of A.mellea strain(99.76%).The DNA sequence of GZDF-1 had the highest similarity with that of A.mellea strain(99.76%).The DNA sequence of strain JLTM had the highest similarity with A.mellea strain(100%).The DNA sequence of strain GZZJ had the highest similarity with that of A.mellea strain(99.88%).Thus,all our strains except AHJZ are A.mellea,and the taxonomic status of AHJZ needs further study because of its low homology with the similar species A.sinapina sequence.
