| Narcissus is a perennial herb which has underground bulb.At present,there are many problems in the production of Chinese Narcissus,due to various factors such as history,source and benifit,Chongming Narcissus(Narcissus tazetta L.var.chinensis)has been disappeared in the flower market for nearly 20 years,it seriously hindered the popularization and application of Chongming Narcissus in China.The study was based on Chongming Narcissus,to study the change law of each inclusion substances in bulbs during storage and their effects on the flower bud differentiation.The bulbs of Chongming narcissus,which were stored at 20℃and room temperature,respectively,so as to provide references for the study of flowering regulation for elucidating the flowering physiology.The main results are shown as follows:1.In the two sets of temperature storage,there was a significant difference in the speed of flower bud differentiation.Observation results showed that,stored for six to seven weeks later,the bulb bud of Chongming Narcissus have completed the whole differentiation process,the stamens,pistils and tepals could be seen very clearly.Relative to room temperature,the bud differentiation process of Chongming Narcissus at 20℃was faster,Compared to room temperature storage,each differentiation stage was about one week to two weeks earlier basically,flower bud differentiation has gone through five periods:inflorescence bud differentiation stage,perianth segment differentiation stage,stamen primordium differentiation stage,pistil primordium differentiation stage,vice crown primordium differentiation stage.2.The changes of starch content and soluble sugar content were evidently,but the change of soluble protein content was not obviously.The results show that starch content increased first and then decreased with prolonged storage time from the absolute in each period,starch content at room temperature were higher than 20℃at the first three weeks,and then significantly lower than 20℃until the end of the observation.Soluble sugar content in bulbs gradually increased at 20℃,the highest value appeared in the 5th week,then declined,and the peak at room temperature appeared in the 2nd week,the highest value was lower than 20℃observed by 40.48%.The change of soluble protein content was between1.812.25mg/g not obviously,and the soluble protein content was found to have a significant increase of 22%five weeks before at 20℃relative to the storage before.Total phenolic content in bulbs decreased in different degrees at room temperature and 20℃,the drop of total phenolic content at20℃was significantly greater than the room temperature.3.The bulbs of Chongming narcissus(Narcissus tazetta var.chinensis),which were stored at 20℃and room temperature,respectively,were used to analyze the level of endogenesis hormones in bulbs for materials,by the method of ELISA(Enzyme-Linked ImmunoSorbent Assay)during storage.The results show that GA content increased gradually up to the 7th week during storage,and the peak at 20℃was higher than room temperature observed by 40.67%.IAA content changes slightly different in the two storage conditions,IAA content at room temperature were higher than 20℃in the 3rd week to the 6th weeks,and the flower bud differentiation process of 20℃was significantly faster than the room temperature in these four weeks.The change of ZR content in room temperature was consistent with the trend of GA,but slightly different changes in 20℃,until the end of the sampling,was still on the rise.ABA content in two groups showed the trend down first and up later,all appeared lowest value in the 3rd week,and the minimum at room temperature was lower than 20℃observed by 16.97%,then 20℃obviously rose faster than room temperature.Therefore,relatively low temperature can accelerate the flower bud differentiation of narcissus bulbs.20℃storage period could promote the narcissus flower bud differentiation,was conductive to the accumulation of GA content,and higher levels of the GA content can accelerate the flower bud differentiation.High levels of ZR has a close relationship with promoting the process of the flower bud differentiation.Endogenous IAA and ABA levels were negatively correlated with the flower bud differentiation.4.Amylase activity at 20℃was higher than the room temperature.POD activity fluctuated slightly in two kinds of temperature treatment,however the overall trend was reduced.The related research results can provide references for the study of flowering regulation,breed of variety,breeding of new variety,etc of Chongming narcissus in the future. |
