Calcium,iron and zinc,as inorganic mineral nutrients,play an important role in the growth and development of plants and animals.Plants as producers are important links for nutrients entering the food chain.Therefore,The quality of forage is very important for the feeding of herbivores.Festuca pratensis is a high quality forage grass that is widely distributed in grassland.Bromus inermis is a high quality forage grass with highly tolerant to low-temperature stress.The root system is developed and the nutritional value is high.In this experiment,the total calcium,iron and zinc content of 105 kinds of Festuca pratensis and 55 kinds Bromus inermis from different countries and regions were determined by Inductively Coupled Plasma(ICP).We also established the transient gene expression methods for these two pastures.The main results are as follows:(1)The soil culture experiment,hydroponic experiment,and petri dish culture were used to determine the content of calcium,iron and zinc,and were screened successively.Finally,the Festuca pratensis high calcium variety 102(ZXY2014p-13032)and the low calcium variety 88(ZXY2014p-12105)were selected,and the high calcium variety calcium content(?g/mg)was about 1.5-3 times of the low calcium variety.At the same time,the variety 15(ZXY2004P-278)with relatively high iron content was screened,which was about 1.5 times that of other varieties.In the roots and leaves of Festuca pratensis,Zinc content has no repeatability.(2)The same method was used to screen the Bromus inermis,and it was found that the variety 46(ZXY2013p-11390)with higher calcium content,it was about 1.5 times of other varieties.There was no obvious repeatability of zinc and iron content in 55 portions of Bromus inermis.(3)In order to analyze the ion absorption capacity of Festuca pratensis,The experiment of the barium absorption rate was then continued.The results show that the absorption capacity of 102 for barium ion greater than that of 88.(4)In order to explore the accumulation of calcium in different leaves of Festuca pratensis,Calcium,iron and zinc contents in old and new leaves of 102 and 88 were determined.The results showed that the calcium content and zinc content was higher in the old leaves of Festuca pratensis.And the calcium content of 102 was always greater than 88.The old leaves of Festuca pratensis did not show higher accumulation of iron content.(5)Finally,the transient expression method of Festuca pratensis and Bromus inermis was established.Successful expression of red fluorescent protein in the endoplasmic reticulum,Golgi apparatus and peroxisome of Festuca pratensis.Successful expression of red fluorescent protein in the peroxisome of Bromus inermis.At present,There are relatively few studies on the content of calcium,iron and zinc in pasture.In this paper,the content of calcium,iron and zinc in 105 kinds of Festuca pratensis and 55 kinds of Bromus inermis from different countries and regions were systematically determined.The results of this study have important value for the evaluation of high quality varieties of Festuca pratensis and Bromus inermis.The establishment of transient transformation methods also laid the foundation for the functional study of related genes.
