Cashmere quality of the Tibetan cashmere goat is higher than other goat breeds,but the quantity of the cashmere can not meet the demands of consumers.Therefore,there is a need to improve the cashmere production in Tibetan cashmere goat breeding industry.The former studies proved that ?-catenin?BMP4?Noggin were the key genes which control the growth of the cashmere,but little is known about Sequence,genetic diversity and mRNA expression of ?-catenin,BMP4 and Noggin in Tibetan cashmere goat.In the present study,210 Tibetan cashmere goats were chose form different counties of Tibet area,and the CDS region of ?-catenin?BMP4?Noggin were obtained.Besides,we also analysed the genetic diversity of the exons of ?-catenin?BMP4?Noggin and the mRNA expression level of these three genes.Results of this study are as follows:1.Length of CDS in ?-catenin?BMP4?Noggin was 2346 bp,1230 bp and 699 bp.These three genes coded 781,409 and 699 amino acids respectively.What's more,similarity of ?-catenin?BMP4?Noggin between the Tibetan cashmere goat and mammals were more than 97.79%.From the phylogenetic tree analysis,the Tibetan cashmere goat,Capra Hircus,Ovi aries and Bovine shared the close relationship while the Tibetan cashmere goat shared the distant relationship with Xenopus laevis and Danio rerio.2.Fourteen coding exons of ?-catenin,three coding exons of BMP4 and one coding exon of Noggin were successfully amplified.With the exon regions sequencing of ?-catenin?BMP4?Noggin,no SNP loci were detected in specific amplified fragment in BMP4?Noggin while five SNP loci were observed in ?-catenin.The heterozygosity of c.621C>T,c.1491C>T,c.63C>T in the five SNP loci were higher while the c.687G>A,c.1140C>T loci were lower.Population genetic analysis revealed that all five SNP loci conformed to Hardy-Weinberg equilibrium law(P>0.05),it means all five SNP loci are stable inheritance,and c.621C>T,c.1491C>T,c.63C>T loci were more research potential.Analysis the heterozygosity of these five SNP loci in six counties with cashmere yield showed that the c.63C>T,c.621C>T,c.687G>A loci may be positively correlated with cashmere yield,while c.1140C>T and c.1491C>T loci may be negatively correlated with cashmere yield.3.mRNA expression level of ?-catenin in Gê'gya cashmere goat was significantly higher than other counties(P<0.05).mRNA expression level of BMP4 in Gêrzê and Gê'gya were also significantly higher than other counties(P<0.05).However,little difference was observed in mRNA expression level of Noggin in Tibetan cashmere goat where from different counties(P>0.05).According to the analysis of the cashmere yield indicate expression of BMP4 may inhibit cashmere growth,but the relationship between the ?-catenin and Noggin with the growth of cashmere is unclear.Of the ?-catenin,different genotypes in cahmere goat may affect the expression level.The above obtained results provide basic data for better understanding about the characrestics of ?-catenin?BMP4 and Noggin of Tibetan cashmere goat,so as to for improving the cashmere production in Tibetan cashmere goat breeding.
