| Muscle fibres are highly diverse in colour,contractile properties and metabolic mechanisms,boriler muscle fiber type is important factor in Modern boriler industry,because muscle fiber type can influence meat quality,metabolism means and efficience of metabolism.Myozenin 3(Myoz3)is one of the candidate genes that may influence muscle fibre type.Since its discovery by three independent groups in the period of 2 years,the Myoz family has emerged as one of the most intensively studied gene families controlling muscle fibre type.The Z-disc plays important roles in both sarcomere structure and signal transduction and is formed by dozens of proteins,including the Myoz family.Myoz1 and Myoz3 are predominantly expressed in fast-twitch skeletal muscle.(1)Chicken Myoz3 was expressed both prenatal and postnatal period,and rise through prenatal phase till the first day after hatch.Higher level of Myoz3 was expressed in breast muscle and leg muscle compared to heart and liver.However,there was no difference of Myoz3 expression was found between species.(2)There were totally 5 coding SNPs were found in 140 experimental chickens,including c.240C>T、 c.501C>T、c.516C>T、c.576C>T and c.675C>G.3 in Avian group and 5 in Yellow bantam group.All SNPs identified in this study were synonymous mutation.In YB group,SNP c.516C>T was significantly associated with L* of breast muscle,haplotype H3 was significanty associated with wings wight ratio,and haplotype H4 was significantly associated with feromal circumference.However,in Avian group,no carcass traits were found to be associated with carcass traits.(3)Under knockdown condition,pathways such as cardiac muscle contraction,ECM-receptor interaction,PPAR,vascular smooth muscle contraction,and focal adhesion were enriched.Key words such as calcium and muscle protein were also enriched.Biological process such as mesenchyme migration,tendon development,long-chain fatty acid import was enriched.Under the overexpression condition,biological process terms such as positive regulation of vascular smooth muscle cell proliferation,loop of Henle development,regulation of energy homeostasis were enriched.Four pathways were found to be enriched,namely,the Erb B signalling pathway,influenza A,herpes simplex infection,and focal adhesion.(4)PPI analysis show that differentially expressed genes encode a highly interconnected network.And further confirmed the results that MYH10 and MYLK2 interact with each other is highly likely molecular process that under Myoz3 regulation.In addition,GATA6 can also bind to MYH10,which is another possible interaction that explain the function of Myoz3.SNPs that were identified in this study are potentially helpful for Marker assistant selection.This is the first study that found the role of ch Myoz3 on CEF proliferation and identified its potential regulation network,which provide valuable evidence for chicken embryonic development research. |
