| Deoxynivalenol(DON)is the most common mycotoxin in feed.DON mediated apoptosis of lymphocytes through oxidative damage,which reduced the immunity of animals.Selenium has the biological characteristics of anti-oxidation and improving the immunity of animals.Cells were stimulated with concanavalin A,a T cell stimulatory lectin,in the absence or presence of DON(100,200,400,800 ng/mL),Se(Na2SeO3,2μmol/L),or combinations of Se2μmol/L and DON 100–800 ng/mL for 12,24,or 48 h.ELISA and qRT-PCR were used to detect the expression of IL-2,IL-4,IL-6,IL-10,IFN-γand IgG,IgM mRNA and protein in porcine spleen lymphocytes.The results and conclusions were as follows:(1)At 12 h,24 h and 48 h,compared with control group,DON exposure significantly and dose dependently decreased the expression levels of IL-2,IL-4,IL-6,IL-10,IFN-γ,IgG and IgM mRNA and protein(P<0.05),except for the individual indicators of D1 groups.It was Shown that DON can cause a decrease in cytokines and immunoglobulins in pig spleen lymphocytes.(2)At 12 h,24 h and 48 h,compared with the control group,Se alone could increase the expression of IL-2,IL-4,IL-6,IL-10,IFN-γ,IgG and IgM mRNA and protein(P<0.05),which indicated that Se could increase the secretion of some cytokines and immunoglobulin.(3)At 12 h,24 h and 48 h,the levels of IL-2,IL-4,IL-6,IL-10,IFN-γ,IgG and IgM protein and gene expression were higher in the selenium and DON groups than in Corresponding to DON alone,but lower than the blank control group.It was shown that selenium can alleviate the decrease of cytokines and immunoglobulin of porcine spleen lymphocytes caused by DON.The results of this investigation demonstrated that Se has the potential to counteract DON-induced immunosuppression in pigs and is a promising treatment for DON-mediated toxicity. |
