The increasingly narrow genetic basis and the shortage of germplasm resources seriously restricted the development of wheat breeding.Psathyrostachys huashanica(2n = 2x = 14,Ns Ns)harbored numerous excellent gene resources and exhibited extensive application prospects as a breeding resources via distant hybridization.The excellent genes can be introduced into the background of common wheat by means of chromosome engineering,so as to increase the diversity of wheat breeding parents and broaden the genetic basis.In this research,H1132,H1133,H4122,H1423,derived from common wheat 7182 and P.huashanica,were characterized by cytological,in situ hybridization,molecular marks and morphological analysis.Results are as follows:(1)Cytogenetic analysis of the offspring derived from common wheat and P.huashanicaThe cytological results of BC1F8 and BC1F9,which derived from common wheat 7182 and P.huashanica,showed that the number of chromosomes ranged from 40 to 54.Among them,the percentage of 42-44 was 82.5%,and the number of 42 chromosomes was 89.the percentage of 45-54 and 40-42 were 10.8% and 6.7% respectively.There was diversity in chromosome configuration of derivatives,a large number of derivatives had univalent,and some derivatives had telocentric chromosome and trivalent.Unstable univalent configurations were still present in 42 and 44 chromosomes.GISH results showed that the chromosome number of alien chromosomes was unstable,and the range was 0-10.(2)Identification and analysis of H1133,H4122 and H1423 derivativesCytological studies showed the chromosome number and configurations of H1133,H4122 and H1423 were 2n = 44 = 22?.The labeled genomic DNA of P.huashanica was GISH probe,Oligo-p Ta535(red)and Oligo-p Sc119.2(green)were FISH probes.GISH-FISH analysis indicated they all had two Ns chromosomes from P.huashanica,and 42 chromosomes from common wheat.three EST(CD454742?BF473348?CD454575)and two PLUG(TNAC1326?TNAC1248)marks,which developed from homoeologous group three,could be used to produce the specific bands of P.huashanica in H1133.one EST(BE499166)and two PLUG(TNAC1588?TNAC1503)marks,which developed from homoeologous group five,could be used to produce the specific bands of P.huashanica in H4122.three EST(BE637663?BG274576?BE591127)and two PLUG(TNAC1957?TNAC1821)marks,which developed from homoeologous group seven,could be used to produce the specific bands of P.huashanica in H1423.So,H1133 was a 3Ns alien addition line,H4122 was a 5Ns alien addition line,H1423 was a 7Ns alien addition line.These materials showed different agronomic characters with 7182,and the average plant height of them were lower than 7182.The tiller of H1423 was less than 7182.At the adult stage,H4122 and H1423 was resistant to stripe rust with the mixture of CYR32 and CYR33.The resistance of H4122 and H1423 originated from P.huashanica.The identification results at seedling stage showed that the resistance of 7182,H1133,H4122 and H1423 to V26,CYR31 and CYR32 was basically the same,both of which were high resistance.H1133(a)and H1133 showed resistance segregation in CYR29 and CYR31.(3)Identification and analysis of H1132 derivativesCytological studies showed the chromosome number and configurations of H1132 was 2n = 42 = 21?.With the labeled genomic DNA of P.huashanica as the GISH probe,Oligo-p Ta535(red)and oligo-p Sc119.2(green)as the FISH probes,GISH-FISH analysis indicated H1132 had two Ns chromosomes from P.huashanica,and 40 chromosomes from common wheat apart from 1D chromosomes.three EST(BE497584?BE443796?BG313767)and three PLUG(TNAC1026?TNAC1043?TNAC1091)marks,which developed from homoeologous group one,could be used to produce the specific bands of P.huashanica in H1132,while Chinese Spring nulli-tetrasomic analysis revealed that wheat 1D chromosomes were absent in H1132.The results were consistent with in situ hybridization analysis.The evaluation of agronomic traits demonstrated that the plant height was significantly lower than 7182,and the resistance originated from P.huashanica.At the adult stage,H1132 was resistant to stripe rust with the mixture of CYR32 and CYR33.The resistance of H1132 originated from P.huashanica.The identification results at the seedling stage showed that H1132 and its parent 7182 showed basically the same performance to V26,CYR31 and CYR32,all of which were immune to high resistance,but affected to CYR29.Above all,H1132 was a 1Ns(1D)alien substitution line,and could be a potentially bridge material in wheat breeding with specific agronomic traits.
