In this study,the buds of‘Korla pear'and‘Dangshansu pear'and their two bud sports were used as materials,and their differences of morphological characteristics,main biological characteristics,quality characteristics and post-harvest storage were compared and evaluated,and the buds were identified using AFLP technology,The research provides a foundation for the identification and evaluation of buds and their application in pear.The main results are as follows:1.According to the testing standard of New plant Cultivar DUS Testing Guideline of Pear,the morphological,biological and quality characteristics of‘Korla pear'and bud sports were compared and analyzed.It was found that there was no significant difference between the floral characters of‘Korla pear'and bud sports.The buds became shorter,the trunk thickness became thinner,the crown diameter became smaller,but the germination rate increased by 23%.There were significant differences between‘Korla pear'and bud sports in single fruit weight,hardness,stone cell and soluble solid content?P<0.05?,bud mutation of the weight of 159.32 g,hardness of 7.51 kg·cm-2,stone cell content of 0.26 g·100g-1,soluble solids content 13.65%;‘Korla pear'fruit weight 147.04 g,hardness of 5.98 kg·cm-2,stone cell content of 0.35 g·100g-1,soluble solids content 12.19%;The content of single fruit weight,hardness and soluble solids were significantly higher than that of‘Korla pear',and the content of stone cells was significantly lower than that of‘Korla pear'.2.According to the testing standard of New plant Cultivar DUS Testing solids content 14.16%;‘Dangshansu pear'weight of 246.96 g,hardness of 5.92 kg·cm-2,stone cell content of 0.39 g·100g-1,soluble solids content 11.85%;The single fruit weight of bud sport was significantly lower than that of‘Dangshansu pear',but the content of hardness and soluble solids was significantly higher than that of‘Dangshansu pear',and the content of stone cells was significantly lower than that of‘Dangshansu pear'.3.Difference comparison and evaluation of different storage methods on fruit quality of‘Korla pear',‘Dangshansu pear'and their two varieties bud sports.The results showed:?1?‘Korla pear'and bud sports were collected and stored at room temperature for 60days.When buds were formed at 30 and 60 days,the hardness decreased by 1.2%and 1.7%,the rise rate of soluble solid content was 10.5%and 3.1%,the titratable acid content decreased by 0%and 27.3%,and the peel turned yellow at 60 days.At 30 d and 60 d,the hardness of‘Korla pear'decreased by 2.8%and 8.9%,and,the rise rate of soluble solid content in 30 days was 4.8%,and the decline rate of 60 days was 0.7%,the decrease rate of titratable acid content was 20%and 20%,and the peel turned yellow at 30 d.?2?‘Korla pear'and bud sports were collected from cold storage for 120 days.When buds were formed at 30 d,60 d and 120 d,the hardness decreased by 3.5%,3.7%and 4.1%,and the rise rate of soluble solid content in 30 d and 60 d is 5.1%and 6.4%,and the decline rate of 120 d is 7.3%,the decrease rate of titratable acid content was 9%,18.2%and 63.6%.At 30 d,60 d and 120 d,the hardness of‘Korla pear'decreased by 7.9%,12.9%and 16.2%,the increase rate of soluble solid content in 30 d was 2.5%,and the decrease rate in 60 d and120 d was 2.1%and 5.3%,and the titratable acid content decreased by 6.7%,40%and 46%.?3?‘Dangshansu pear'and bud sports were collected and stored at room temperature for60 days.When buds were formed at 30 and 60 days,the hardness decreased by 4.8%and6.7%,the rise rate of soluble solid content was 2.6%and 3.2%,and the titratable acid content decreased by 8.3%and 8.3%.At 30 d and 60 d,the hardness of‘Dangshansu pear'decreased by 12.3%and 13.7%,the increase rate of soluble solid content was 7.9%and12.7%respectively,and the soluble acid content decreased by 7.7%and 7.7%.?4?‘Dangshansu pear'and bud sports were collected and stored in cold storage for 120days.When buds were formed at 30 d,60 d and 120 d,the hardness decreased by 1.8%,2.2%and 3.6%,the increase rate of soluble solid content in 30 d was 4.2%,and the decrease rate of 60 d and 120 d was 0.1%and 1.5%,and the content of titrable acid decreased by8.3%,41.7%and 41.7%.At 30 d,60 d and 120 d,the hardness of‘Dangshansu pear'decreased by 1.9%,3.5%and 4.4%,the rise rate of soluble solids in 30 d and 60 d was 2.1%and 0.3%,and the decrease rate of 120 d was 0.4%,and the content of titrable acid decreased by 0%,38.5%and 38.5%.These results showed that the bud sports of‘Korla pear'is more resistant to storage than that of‘Korla pear',while the bud sports of‘Dangshansu pear'and‘Dangshansu pear'have little difference in storage resistance.4.AFLP technology was used to identify buds and verify the genetic difference between‘Korla pear',‘Dangshansu pear'and their corresponding bud sports.The results showed that the amplification bands were different in both quality?with or without fragments?and quantity?strength and weakness of fragments?,which indicated that there were differences in DNA level between the budding variety and the original variety.There were 91 distinct spectral bands and 86 distinct spectral bands in‘Korla pear'and bud sport,a total of 84spectral bands,9 polymorphic bands,polymorphic percentage was 9.68%,the genetic similarity coefficient was 0.9492,and the genetic distance was 0.0508.‘Dangshansu pear'and bud sport have 90 and 86 distinct spectral bands respectively,including 85 spectral bands,6 polymorphic bands,polymorphic percentage of 6.59%,genetic similarity coefficient of 0.9659,genetic distance of 0.0341.
