Amur Tiger?Panthera Tigris altaica?is an endangered carnivore that is now listed on the IUCN red lis.The Amur Tiger is an endangered carnivore that has been listed in the IUCN Red List of Endangered Species.It is mainly distributed in the Far East of Russia,the Northeast of China and the northern part of North Korea.In recent years,the population of wild Amur tigers has increased,but the situation is still not optimistic.In the tigers,natural habitats,tigers and humans face serious conflicts in resources,and the number of tigers is largely related to human activities,for the maintenance of the tiger population,on the one hand it is very important to protect the environment on which to survive.On the other hand,the zoo's protective research also plays a vital role.This paper uses the modern scientific method to study the relationship between blood physiological parameters,blood microbiota composition and intestinal microbes of different ages.The main research contents are as follows:?1?Measurement of blood physiological parameters of captive Amur tigers.In this study,blood physiological parameters of 26 captive Amur tigers were measured by hematology analyzer,and the differences in physiological parameters between male and female individuals were compared,and platelet?PLT?related parameters and effects were calculated.The results showed that there was no significant difference in blood physiological parameters between male and female tigers?P>0.05?.Plt-related parameters,PLT 211.15±59.77 109·L-1,PCT 0.26±0.09%,MPV 12.65±0.88 fL,PDW 22.28±1.68 fL,P-LCR44.56±8.03%were listed.The determination of blood physiological parameters provides great help for the diagnosis and treatment of the Amur tiger diseases,and the determination of plt-related parameters provides a scope for the establishment of standardization of blood physiological parameters of the Amur tiger.?2?Study on microbial composition of blood of captive amur tigersThis experiment aims to study the blood bacterial composition of Amur tiger in captive conditions.Six?three males and three females?healthy captive Amur tigers were selected,and the microbial genomic DNA was extracted from the blood of the animals using sterile equipment.And analysis of bacterial composition in blood using 16S high-throughput technology sequencing.The results showed that the dominant bacteria in the blood of Amur tiger were Proteobacteria?46.12-56.24%?,Firmicutes?30.82-40.18%?,Bacteroidetes?7.6-13.57%?,Actinobacteria?0.15-2.19%?and Cyanobacteria?0.54-0.95%?.The dominant microorganisms in the horizontal order are Staphylococcus?33.87%?,Caulobacteraceaenorank?23.32%?,Raoultella?8.58%?,Bradyrhizobium?6.75%?,Sediminbacterium?5.91%?,Acinetobacter?4.67%?and Asinibacterium?4.51%?.There was no significant difference in blood microbial composition between male and female animals.This experiment basically confirmed the presence of microorganisms in the blood of healthy captive Amur tiger by 16S high-throughput technology,which is of great significance for the health protection and disease prevention of Amur tiger.?3?Analysis of fecal microflora diversity of amur tigers of different ages.This experiment aims to study the bacterial abundance and diversity of Siberian tiger feces under different ages.Nine of the Siberian tigers were selected and divided into three groups:larvae,sub-adults and adult.After collecting fresh feces,16S high-throughput sequencing was performed to analyze the abundance and diversity of the gut microbes in the Northeast Tiger.The results showed that the dominant bacteria in the feces of Siberian tiger were Firmicutes,Actinobacteria,Bacteroidetes,Fusobacteria and Proteobacteria.The most dominant bacteria in the intestinal tract of group adult were Firmicutes?73.93%?,the second dominant bacterium was Actinobacteria?16.09%?;the most dominant bacterium in the sub adult group was Firmicutes?50.97%?,and the second dominant bacterium was Actinobacteria?21.36%?;The most dominant bacteria in the childhood group were Firmicutes?58.90%?,and the second dominant bacteria was Bacteroidetes?18.92%?.OTU profiles of abundance and diversity of the major genera were assessed.This experiment analyzes the relationship of fecal microbes in different age groups by 16S high-throughput technology,which is of great significance for the health protection and disease prevention of Amur tiger.
