Agriculture develops very fast in our country with a broad territory.Alfalfa is the king of forage because of its rich nutrition and strong adaptability to the environment.Along with the change of farming mode,the application and usage of herbicides have became more and more widely.Variety improvement of alfalfa is the key to reduce the impact of herbicides on the growth of it.Although conventional breeding methods may improve the resistance of alfalfa,but the less plant area and longer breeding time become the bottleneck of it.Therefore,it is important to find an efficient and convenient method which is herbicide-resistant transgenic alfalfa and male sterile line hybridization method.To shorten the breeding period of breeding homozygous offspring and greatly improve the efficiency of F1 hybrid seed production,we take the male sterile line of alfalfa as the female parent.1.We select the candidate glufosinate-resistant alfalfa as materials of bar gene in this experiment,and detect the exogenous genes of the candidate materials by bar test strip and PCR method,to finally confirm a total of 15 positive plants.In addition,with herbicide smearing method the critical concentration of 7‰ was determined,which could distinguish resistant alfalfa from non-resistant alfalfa.After that,11 plants with resistance of glufosinate positive strains were determined by herbicide spraying method.2.11 hybrid combinations were allocated with 11 herbicide-resistant transgenic alfalfa plants as male parent and male sterile line as female parent and take Gongnong 1 as the control variety.The experiment tested the offspring for molecular and herbicide resistance,along with evaluation and selection of yield and quality performance,from which 7 hybrid combinations with excellent performance were selected.To sum up we know that the method of using herbicide-resistant transgenic alfalfa male parent to cross with sterile line female parent is feasible.We can effectively solve the problem of weed control in alfalfa production due to the selection of good herbicide-resistant transgenic materials,in return it provides more material basis for the promotion of forage industry.
