Potato?Solanum tuberosum L.?is an important tuber crop in the world,and China is the largest producer and consumer of potatoes.The potato is comprehensive nutrition and rich in starch,vitamins,carotenoids,anthocyanins and a variety of mineral elements,It play an important role in national food security and the nutritional health of human.Anthocyanins are the main pigment components of colored potatoes and are also important nutrients.It makes the breeding of colored potatoes being attracted much attention in recent years.However,the genes regulating the color of potato tuber flesh have not been analyzed,which seriously restricted the molecular breeding and the analysis of the metabolic network.Major genes controlling anthocyanin synthesis by population mapping will provide molecular markers for colored potato breeding and accelerate the progress of potato molecular breeding.The analysis of gene functions will lay a foundation for the study of regulatory network of potato flesh pigment accumulation.Two BC1S1 segregation populations were used:purple/red/yellow and red/yellow segregation population.The following results were obtained by mapping the genes involved in the regulation of anthocyanin synthesis:?1?BSA initial localization,fine mapping,candidate gene prediction analysis were performed on purple/red/yellow segregation population,The gene controlling the accumulation of purple anthocyanin in tuber flesh was located at the end of chromosome 10,and the candidate genes were predicted to be PGSC0003DMG400013966 and PGSC0003DMG400013965;?2?BSA preliminary mapping,fine mapping,predictive analysis of candidate genes,protein sequence alignment and expression analysis were performed in red/yellow segregation population.PGSC0003DMG400013965 was determined to be the I gene that regulating accumulation of anthocyanin in tuber skin,and it was speculated that could regulate the accumulation of anthocyanin in potato tuber flesh and skin as a whole.It is known that the genes controlling the red appearance of tuber flesh were located at the end of chromosome 10.The candidate genesareconfirmedtobePGSC0003DMG400013966and PGSC0003DMG400013965;?3?The genetic transformation experiments of PGSC0003DMG400013966 and PGSC0003DMG400013965 genes were carried out.Four positive plants with mutation of PGSC0003DMG400013966 gene were obtained,and no positive plants were obtained for the PGSC0003DMG400013965...
