| In recent years,goose breeding industry has become a sunrise industry of China’s aquaculture industry and has a good development pros PFct.TThere are many factors restricting the development of goose breeding and its sideline,among which seasonal laying of goose is one of the key factors.Therefore,it is necessary to study the development of goose follicles so as to better promote the development of goose breeding industry.Now,some scholars have found that the ERK signaling pathway can regulate the pre-level follicular development of poultry through some pathways.The pre-follicles of the seed goose were collected and used as test samples.In this study,q PCR,Western Blot and IHC techniques were used to detect the expression of target genes,m RNA and protein levels,and the expression sites in the pre-follicles of the target gene goose.The research results are as follows:(1)q PCR assay technique was used to study the m RNA expression levels of key genes in the ERK signaling pathway in the pre-follicles of the geese,and to test their significance.The results showed that all target genes were expressed in the five pre-follicles of the goose and there was a significant difference in the data results(p < 0.05).The expression patterns of Raf1,Myc and Mek1 genes firstly increased,then decreased,then increased and then decreased.The target gene Raf1 and Myc were the highest in LWF,and the target gene Mek1 was highest in SWF.The expression patterns of the target genes Hras,Mos and ERK showed a continuous increase trend,and the expression level in PF follicles was the highest.The expression pattern of the target gene Srf showed a "W" trend of decreasing first,then increasing and then decreasing,and its expression level was the highest in SYF.(2)The results of qPCR were analyzed by correlation analysis.The results showed that Hs,Mek1,ERK and Mos genes play a major role in the regulation of ERK signaling pathway in PF follicles,small white follicles and medium white follicles,but in LWF and SYF.The Raf1,Myc and Srf genes facilitate the regulation of follicular development by the ERK signaling pathway.(3)The expression sites of the target proteins Hras,Mek1 and ERK in the pre-follicles of the goose were analyzed by immunohistochemistry.The results showed that expression sites were detected in both granular layer cells(GC)and membrane cells(TC)in the five pre-follicles of the goose,and the results were positive.(4)The protein expression levels of the target genes Hras,Mek1 and ERK in the anterior follicles of the goose were studied by Western blotting.Among them,the target protein ERK was the highest in SWF and the lowest in SYF(P <0.05).The target protein Hras expressed the highest amount in SWF and the lowest expression in LWF(P<0.05).The target protein Mek1 had the highest expression in PF and the lowest expression in MWF(P < 0.05).(5)Based on the gene expression and follicular development of ERK signaling pathway,we can find that ERK signaling pathway can promote follicular development in pre-grade seeded geese. |
