The occurrence of jujube black spot disease in southern Xinjiang has seriously hindered the sustainable and healthy development of the jujube industry.At present,the control of this disease by many scholars mainly focuses on the morphological structure and physiological and biochemical properties of the pathogenic bacteria,and there are few reports from the molecular level.Therefore,this paper attempts to investigate and analyze jujube after the induction of Alternaria altrenata,differential changes at the transcriptome level to uncover functional genes associated with jujube black spot resistance from a molecular perspective,the regulation of key genes in jujube fruit lays a molecular theoretical foundation for the control of this disease.1.By exploring the main pathogenic bacteria that cause jujube black spot disease in southern Xinjiang,it will lay the foundation for the accurate screening of subsequent disease resistance genes and target control.In this paper,a combination of high-throughput sequencing techniques and traditional isolation and identification methods were used to conduct statistical analysis of disease incidence,tissue isolation,morphological observation,molecular biology analysis and pathological inoculation tests on diseased fruits from Kashi,Hetian and Aksu regions using gray jujube and junzao fruits as test materials,A.alternata was identified as the main causative agent of jujube black spot in the south;Junzao field prevalence is 9.4 times higher than gray jujube,gray jujube is more resistant to disease than junzao.2.To study jujube fruit induction in A.alternata at the molecular level of the response mechanism to screen for disease resistance genes.Analysis of differential gene expression and associated metabolic pathways based on RNA-seq assays of gray jujube and junzao fruits infested with A.alternata for 0-4d;The results showed that most of the genes in junzao showed expression differences,and these differential genes were mainly distributed in metabolic pathways such as plant hormones,flavonoid biosynthesis and glutathione;It is speculated that the method of junzao to resist the invasion of foreign pathogens can be achieved by regulating the expression of important genes in metabolic pathways such as plant hormones,flavonoid biosynthesis and glutathione.3.In order to further explore the differential gene expression,select the six genes with the most differences from the gray jujube treatment group: expansin-A1,GMP synthase,glucose-6-phosphate/phosphate translocator 2,Amino acid permease,PTR FAMILY 5.4 and sulfate transporter 3.4 and verify them by qRT-PCR;The results showed that qRT-PCR detection results were consistent with Illumina sequencing results;The expression levels of 6 disease resistance-related genes changed significantly with the extension of infection time,it is speculated that the change in expression level is a kind of defense response of plant fruits.
