Berberine is a main ingredient in the Coptis,one of the common traditonal medicines,which can be extracted from natural medicinal herbs such as golden cypress,barberry,and root of mahonia bealei.Berberine has broad-spectrum antibacterial properties,which can effectively inhibit G?and G?bacteria in vivo and in vitro,such as escherichia coli,shigella dysenteriae,staphylococcus aureus,bacillus subtilis,etc.In addition,Berberine is used in the treatment of a wide range of bacterial diseases of livestock and poultry.This experiment investigated the distribution of different doses of berberine in white feather broiler tissues and the distribution change of berberine after absorption in the tissue,after the administration of compatible borneol.To provide theoretical basis for the research and development of berberine pharmaceutical preparations and rational use of clinical drugs.The experiment used white feather broilers as test subjects.By oral administration of different doses of berberine and combination group?berberine+borneol?,sacrificed at four time points:2,4,8,12 h.simultaneously,quickly dissect,take the tissue.The content of berberine in tissue samples was determined by HPLC.The established liquid chromatography detection condition in this experiment is:detection wavelength of 346 nm.The mobile phase was:phosphate buffer[?0.05 mol/L sodium heptane sulfonate:0.05 mol/L potassium dihydrogen phosphate?1:1?,0.2%triethylamine was added,and the pH was adjusted to 3.0±0.1 with phosphoric acid?]:acetonitrile=68:32?v/v?.Palmatine was used as the internal standard.Using blank tissue as a standard curve,berberine has a good linear relationship in the range of 0.055 mg/kg,the LOD is 0.05 mg/kg.The average extraction recovery rate of berberine was more than 80%,the RSD was less than 15%.The data proves that the detection method has excellent precision and accuracy,compliance with biological sample determination,and meets the requirements of drug detection and analysis.By measuring the concentration of berberine in the three groups of low,medium and high doses?50 mg/kg,100 mg/kg,200 mg/kg?after intragastric administration in white feather broiler chickens,the collected data were analyzed and processed.It shows that the berberine concentration in each tissue is liver,kidney,heart,pancreas,spleen,lung,fat,pectorales,leg muscle,brain.With the increase of dose,the drug concentration in tissues all increased in a dose-dependent manner.One-way analysis of variance was used to calculate the drug concentration of the three dose groups in heart tissue,the difference was significantly?P<0.05?.Compared with medium-dose berberine when administered with borneol?100 mg/kg+50 mg/kg?,the content of berberine in each tissue was determined,the concentrations of combination group?berberine+borneol?were increased in all tissues compared with berberine alone.The data of two groups were analyzed by t-test.Combined with borneol,the drug Cmaxax in liver and heart was?285.61±79.28?mg/kg and?46.85±8.29?mg/kg,t-test results showed that the difference was significant?P<0.01?.The drug Cmaxax in lung was?6.22±1.05?mg/kg,the difference was significant?P<0.05?.The ratio of berberine peak concentration of each tissue in the compatible with borneol group to the medium dose berberine group was calculated.The Ce values were all greater than 1,which proved that borneol could effectively promote the tissues distribution of berberine.The Ce value was the highest in the kidney,which indicated the tendency of the drug was better in the kidney.This experiment proves that berberine is widely distributed in white feather broiler tissues.Borneol can promote the absorption and distribution of berberine in the tissues,provide a reference for the formulation of drug administration program and the development of new drug preparations.
