In this study,51 Actinidia arguta resources were used as materials,and pseudomonas syringae,the pathogenic bacterium of canker was used Pseudomonas syringae pv.actinidae m228 was used to inoculate the stems and leaves of Actinidia arguta in vitro and evaluate the resistance to canker disease.To establish the evaluation method of Pseudomonas syringae pv.actinidae of Actinidia arguta and determine the resistance to canker disease of different resources of Actinidia arguta.At the same time,the activities of defense enzymes and the changes of secondary metabolites of ‘Kuilv',‘Hongyang' and ‘Xuxiang' were measured before and after inoculation,in order to clarify the physiological mechanism of resistance to canker.The main results are as follows:1.The results of inoculation of semi lignified in vitro stems in different years were basically the same and reached a very significant correlation level,the correlation coefficient was 0.9359.The inoculation of semi lignified in vitro stems could be used as a method to identify the resistance to pseudomonas syringae pv.actinidae of Actinidia arguta;2.The susceptible degree of Actinidia arguta germplasm resources couble be divided into five levels: high resistance,medium resistance,susceptible,medium sensitivity and high sensitivity.Of the 51 germplasm resources evaluated,33 were high resistance resources,18 were medium resistance resources,and there were no susceptible,medium sensitive and high sensitive germplasm resources.3.After inoculation,the increase of defense enzyme activity of Actinidia arguta ‘Kuilv' was higher or significantly higher than that Actinidia chinensis Planch ‘Hongyang' and Actinidia chinensis var.deliciosa ‘Xuxiang'(P<0.05);the change of PPO activity was the largest,which reached the peak earlier than other enzyme activities,indicating that it played an inhibitory role in the early stage of canker disease;it was considered that the activity of SOD,CAT,POD,PAL and PPO could be used as physiological and biochemical index to reflect the resistance of kiwifruit material to pseudomonas syringae pv.actinidae.4.Six kinds of secondary metabolites were detected before and after inoculation for ‘Kuilv' and ‘Xuxiang',and five kinds of secondary metabolites were detected before and after inoculation for ‘Hongyang'.With the increase of inoculation time,the content of secondary metabolites of ‘Kuilv' increased,and the increasing range was larger than ‘Xuxiang'.After inoculation,the secondary metabolites of the seed ‘Hongyang' either decreased or had no obvious change.5.The contents of chlorogenic acid,caffeic acid,rutin and ferulic acid in the six secondary metabolites determined by HPLC were closely related to the mechanism of Actinidia arguta,which were important defensive substance against the invasion of pseudomonas syringae pv.actinidae.
